New advance in the application of compound-specific isotope analysis (CSIA) in identifying sources, transformation mechanisms and metabolism of brominated organic compounds

Summary: This article introduces the current analytical methods for measuring the stable isotopes of carbon, hydrogen, oxygen, and bromine in BOCs using mass spectrometry. It discusses the applications for identifying sources, tracing transformation pathways, and biological metabolisms, as well as the challenges and prospects regarding bromine isotope effects, multiple isotope enrichment, and position-specific isotope analysis. It also highlights the future activities that may benefit the development of the fast-growing field of CSIA.

Using Physical Organic Chemistry Knowledge to Predict Unusual Metabolites of Synthetic Phenolic Antioxidants by Cytochrome P450

Summary: Biotransformation plays a crucial role in regulating the toxicity of organic compounds in organisms. However, the understanding of biotransformation for emerging pollutants is limited due to their unusual reactions. This study used concepts from physical organic chemistry to reveal the unusual metabolic mechanism of synthetic phenolic antioxidants catalyzed by CYP450. Two unusual metabolites were predicted and confirmed through in vitro analysis. The study highlights the importance of physical organic chemistry in predicting the fate of unknown synthesized organic compounds in the future.

Precision Biotransformation of Emerging Pollutants by Human Cytochrome P450 Using Computational-Experimental Synergy: A Case Study of Tris(1,3-dichloro-2-propyl) Phosphate

Summary: Precision biotransformation is a strategy that aims to elucidate detailed biotransformation pathways of complex compounds, such as TDCIPP, by combining high-accuracy quantum chemistry, in vitro experiments, and targeted computations. This study demonstrates the successful mapping of TDCIPP metabolism by human CYP450 enzymes through a three-step synergy strategy, providing insights into the relative yields of different metabolites and proposing a new mechanism involving oxidation and reduction functions of CYP450 enzymes. The proposed strategy shows promise in meeting the challenges of understanding biotransformation mechanisms of synthesized organic compounds in the future.
