Mitochondrial-derived vesicles in skeletal muscle remodeling and adaptation

Anna Picca et al.

Summary: Mitochondrial remodeling is critical for meeting the energy demands of muscle contraction and regeneration. Mitochondrial quality control processes, including biogenesis, dynamics, and mitophagy, are essential for maintaining a functional mitochondrial network and supporting muscle regeneration. Alterations in these processes can lead to impaired muscle development, defective regeneration, and loss of muscle function. Among these processes, mitophagy plays a major role in removing dysfunctional mitochondria through interaction with the endo-lysosomal system. Additionally, mitochondrial-derived vesicles (MDVs) have been identified as a complementary pathway for mitochondrial disposal and potential signaling at the systemic level. This article discusses the generation and release of MDVs as a mechanism for maintaining mitochondrial homeostasis in skeletal muscle and their potential role in immune signaling during muscle remodeling and adaptation.

Isolation of mitochondria-derived mitovesicles and subpopulations of microvesicles and exosomes from brain tissues

Pasquale D'Acunzo et al.

Summary: This article provides a detailed protocol for isolating extracellular vesicles (EVs) from murine and human brains, as well as downstream protocols for characterizing and analyzing these EVs. The technique allows for quantitative and highly reproducible analysis of EV subtypes in normal physiological processes and pathological brain conditions.

MIROs and DRP1 drive mitochondrial-derived vesicle biogenesis and promote quality control

Tim Koenig et al.

Summary: The study characterizes a MIRO1/2- and DRP1-dependent MDV biogenesis pathway and reveals how MDVs shuttle fully assembled protein complexes to lysosomes, maintaining mitochondrial proteome quality. This provides insights into a selective quality control mechanism for multi-subunit complexes within mitochondria.

Could Extracellular Vesicles Contribute to Generation or Awakening of Sleepy Metastatic Niches?

Alberto Hernandez-Barranco et al.

Summary: Pre-metastatic niches provide favorable conditions for tumor cells, with tumor-derived extracellular vesicles potentially playing a role in early metastatic dissemination and dormancy. Further research is needed to confirm the molecular mechanisms of extracellular vesicles in the formation of metastatic niches and tumor cell awakening.

Selective packaging of mitochondrial proteins into extracellular vesicles prevents the release of mitochondrial DAMPs

Kiran Todkar et al.

Summary: Mitochondrial content in extracellular vesicles (EVs) can exacerbate inflammation, while its role in non-inflammatory conditions remains unclear. The research demonstrates that mitochondria-derived vesicles target material to EVs, while Parkinson's disease-related protein Parkin directs damaged mitochondrial content to lysosomes, shedding light on mitochondria-driven immune responses.
