Summary: Polyphosphate (polyP) plays diverse roles in cells from bacteria to humans. This study investigates the mechanisms underlying the toxicity of polyP accumulation outside lysosome-like organelles in yeast. Label-free mass spectrometry reveals that the ectopic expression of bacterial polyphosphate kinase (PPK) activates a stress response mediated by the Yak1 and Hog1 kinases and the Msn2 and Msn4 transcription factors. The toxicity is countered by a pair of polyphosphatases, Ddp1 and Ppx1, that degrade polyP. These findings provide insights into the molecular processes and mechanisms of polyP toxicity.
