Highly Sensitive and Specific Responses of Oyster Hemocytes to Copper Exposure: Single-Cell Transcriptomic Analysis of Different Cell Populations

Jie Meng et al.

Summary: In this study, the responses of different cell populations of hemocytes in oysters under copper exposure were investigated using single-cell transcriptome profiling. The results showed that different cell types had heterogeneous responses to copper, with the granulocyte being the most important responsive cell type and displaying heterogeneity responses of its two subtypes. These findings have significant implications for understanding the impact of copper contamination on oysters.

Single-Cell Ribonucleic Acid Sequencing Clarifies Cold Tolerance Mechanisms in the Pacific White Shrimp (Litopenaeus Vannamei)

Weilin Zhu et al.

Summary: In this study, the molecular mechanisms underlying cold tolerance in Litopenaeus vannamei were preliminarily elucidated through single-cell RNA sequencing analysis. By comparing hepatopancreas cells at different temperatures, differentially expressed genes associated with the immune system, cytoskeleton, antioxidant system, digestive enzyme, and detoxification were identified between cold-tolerant and common types. Additionally, two distinct fates of hepatopancreas cells under low temperature stress were discovered.

The Mechanism of Carbonate Alkalinity Exposure on Juvenile Exopalaemon carinicauda With the Transcriptome and MicroRNA Analysis

Kunpeng Shi et al.

Summary: This study investigated the alkalinity-adaptation mechanisms of the ridge tail white prawn by analyzing mRNA and miRNA transcriptomes. The results showed that ATP binding protein, carbonic anhydrase, and up-regulated miRNAs played important roles in adapting to alkalinity, while immune-related genes protected the prawns from bacterial infection. These findings contribute to a better understanding of the adaptation mechanisms of economic aquatic animals to alkaline environments and provide a theoretical basis for future breeding efforts.

Integrated application of multi-omics approach and biochemical assays provides insights into physiological responses to saline-alkaline stress in the gills of crucian carp (Carassius auratus)

Yingjie Liu et al.

Summary: This study reveals the molecular and metabolic mechanisms of crucian carp under saline-alkaline stress, demonstrating that long-term high saline-alkaline stress causes gill oxidative stress and changes in physiological parameters. Through multi-omics and biochemical analysis, the research found that crucian carp regulates ammonia transport through Rh glycoprotein, increases glutamine and urea synthesis to mitigate the adverse effects of saline-alkaline water, and induces apoptosis and immune response.

Single-cell transcriptomic analysis of neuroepithelial cells and other cell types of the gills of zebrafish (Danio rerio) exposed to hypoxia

Wen Pan et al.

Summary: This study conducted a comprehensive transcriptomic analysis of zebrafish gill cells using single-cell RNA sequencing. The analysis revealed 16 cell clusters, including NECs, neurons, pavement cells, endothelial cells, and mitochondrion-rich cells. NECs in a hypoxic environment showed differential gene expression related to cell cycle control and proliferation. This research provides a foundation for future studies on the molecular biology and physiology of the gill organ.

The effects of salinities stress on histopathological changes, serum biochemical index, non-specific immune and transcriptome analysis in red swamp crayfish Procambarus clarkii

Yi Xiao et al.

Summary: This study investigated the effects of different salinity stresses on the histoarchitecture, serum biochemical indices, antioxidant responses, and transcriptome analysis of Procambarus clarkii. The results showed that increasing salinity led to increased damage to the nuclei and mitochondria, enhanced gill wrinkles, altered serum biochemical indices, decreased antioxidant capacity, and changes in immune-related gene expression. These findings contribute to our understanding of the response of farmed organisms to salinity stress and have implications for the cultivation of P. clarkii in saline conditions.

Cell type diversity in scallop adductor muscles revealed by single-cell RNA-Seq

Xiujun Sun et al.

Summary: This study used single-cell RNA sequencing to explore the molecular features of various cell types in scallop muscles, identifying 20 cell clusters mainly classified into mesenchymal stem cells, muscle cells, neurons, and haemolymph cells. The research revealed a diverse repertoire of neurons in the striated adductor muscle, but not in the smooth muscle, and reconstructed cell differentiation events using pseudotemporal trajectories. The spatial distribution of mesenchymal stem cells and neurons in scallop adductor muscles was also visualized through immunodetection and electron microscopy observation.

Tissue and salinity specific Na+/Cl- cotransporter (NCC) orthologues involved in the adaptive osmoregulation of sea lamprey (Petromyzon marinus)

A. Barany et al.

Summary: Two orthologues of the gene encoding the Na+-Cl- cotransporter (NCC), termed ncca and nccb, were found in the sea lamprey genome. The NCCs occupied basal positions within the NCC clades in a phylogenetic comparison among other vertebrate NCC and NKCC sequences. nccb is likely the key ion cotransport protein for ion uptake by the lamprey intestine that facilitates water absorption in seawater.

Intracellular pH regulation: characterization and functional investigation of H+ transporters in Stylophora pistillata

Laura Capasso et al.

Summary: This study provides the first comprehensive characterization of H+ transporters that may contribute to the homeostatic acid-base balance in coral cells, including members of the Na+/H+ exchanger, vacuolar-type electrogenic H+-ATP hydrolase, and voltage-gated proton channel families, as well as a novel member of the HvCN gene family. Key residues contributing to substrate specificity, protein function and regulation were identified, along with tissue-specific expression patterns and minimal impact of seawater acidification on most of these proteins.
