Collateral sensitivity to pleuromutilins in vancomycin-resistant Enterococcus faecium

Summary: Acquisition of resistance to one antibiotic can sometimes increase susceptibility to a second antibiotic. In this study, it was found that vancomycin-resistant Enterococcus faecium strains showed remarkable increase in susceptibility to pleuromutilin antibiotics (such as lefamulin). The trade-off between vancomycin and pleuromutilins is mediated by epistasis between van gene cluster and msrC, suggesting pleuromutilins may be useful in treating vancomycin-resistant E. faecium infections.

Proteomics Study of the Synergistic Killing of Tigecycline in Combination With Aminoglycosides Against Carbapenem-Resistant Klebsiella pneumoniae

Summary: This study used proteomics analysis to investigate the impact and mechanism of tigecycline and aminoglycoside combination therapy on carbapenem-resistant Klebsiella pneumoniae (CRKP). The study found that the proteomic responses to tigecycline and aminoglycosides were different in monotherapy, with tigecycline dominating the proteomic alterations in combination therapy. Adaptive responses to tigecycline were associated with the upregulation of oxidative phosphorylation and translation-related proteins, while tigecycline might perturb adaptive responses to aminoglycosides through inhibition of heat shock response.

In vitro and in vivo synergistic effects of tigecycline combined with aminoglycosides on carbapenem-resistant Klebsiella pneumoniae

Summary: The study found synergistic or partial synergistic activity between tigecycline and aminoglycosides against CR-KP. Combination therapies showed better therapeutic effectiveness compared to monotherapy for susceptible isolates, indicating in vivo synergistic effects. Additionally, the combination of tigecycline with an aminoglycoside significantly reduced the occurrence of tigecycline-resistant mutants.

The adapted strains showed decreased susceptibility to some antibiotics and increased resistance to others, providing valuable information for designing effective antibiotic regimens against A. baumannii infections.
