Ocular application of electrospun materials for drug delivery and cellular therapies

Development and evaluation of polycaprolactone-based electrospun nanofibers containing timolol maleate as a sustained-release device for treatment of glaucoma: in vivo evaluation in equine eye

Summary: This study developed nanofibrous inserts for sustained ophthalmic delivery of timolol maleate (TIM) for the treatment of glaucoma. The inserts showed suitable physicochemical properties and stability for ophthalmic administration, and demonstrated prolonged release of drug in vitro for more than 3 days. In vivo evaluation showed that the inserts were non-irritant and non-toxic to equine eyes, while effectively reducing intraocular pressure (IOP) for 6 days. These findings suggest that the prepared nanofibrous inserts could enhance patient compliance by reducing the frequency of administration in the treatment of glaucoma.

Delivery of Therapeutics from Layer-by-Layer Electrospun Nanofiber Matrix for Wound Healing: An Update

Summary: The increasing incidences of chronic wounds have led to the demand for development of effective therapeutic wound treatment. Electrospun nanofiber has been recognized as a better system for wound dressing, with the proposed multilayer structure aiming to address the issues of burst release effect in single-layer designs. This review highlights the potential of layer-by-layer multilayer electrospun nanofiber as a superior strategy for designing optimal wound dressings.

The pro-inflammatory response of macrophages regulated by acid degradation products of poly(lactide-co-glycolide) nanoparticles

Summary: This study investigates the immunological effects of PLGA degradation products on cell viability, apoptosis, polarization, and inflammatory reaction. The results show that PLGA degradation induces an acid environment that decreases cell viability and increases cell apoptosis, impacting cell function. The degradation products of PLGA can also convert macrophages to a pro-inflammatory state, providing insight into PLGA-induced inflammation for the design of future biomaterials.

Biocompatibility Study of Electrospun Nanocomposite Membranes Based on Chitosan/Polyvinyl Alcohol/Oxidized Carbon Nano-Onions

Summary: This study synthesized four different nanofibrous membranes of chitosan (CS)/(PVA)/oxidized carbon nano-onions (ox-CNO) using the electrospinning method and conducted subdermal implantations in Wistar rats. The results showed that the nanofibers were continuous, with narrow diameters, and the introduction of ox-CNO increased the crystallinity index, enhancing the material's performance, with good biocompatibility as evidenced by the subdermal implantations.

Degradation and Characterisation of Electrospun Polycaprolactone (PCL) and Poly(lactic-co-glycolic acid) (PLGA) Scaffolds for Vascular Tissue Engineering

Summary: The study evaluated the characteristics and degradation effects of PLGA- and PCL-based nanofibrous scaffolds, finding that longer electrospinning times resulted in stronger mechanical properties and maintained fiber structure. Both PCL and PLGA scaffolds demonstrated adequate degradation over time, with slight increase in fiber diameter and substantial decrease in pore size and porosity percentage. This experiment suggests promise for PCL and PLGA in tissue engineering and regenerative medicine applications.
