Synthesis of new diphenyl urea-clubbed imine analogs and its Implications in diabetic management through in vitro and in silico approaches

Summary: This study synthesized Schiff bases of 1,3-dipheny urea and evaluated their anti-diabetic capability by inhibiting alpha-glucosidase. The compounds showed potent inhibition with IC50 values ranging from 2.14-115 mu M. Docking results indicated the important role of Glu277 and Asn350 in stabilizing these compounds at the active site of the enzyme. These molecules exhibited promising pharmacokinetics, physicochemical properties, and drug-likeness profile. The anti-diabetic potential of these compounds highlights their medical importance and potential therapeutic options for treating type II diabetes.

Synthesis and evaluation of novel xanthene-based thiazoles as potential antidiabetic agents

Summary: A series of xanthene-based thiazoles were synthesized and characterized. These compounds exhibited high inhibition potential against acetylcholinesterase, butyrylcholinesterase, alpha-amylase, and alpha-glycosidase enzymes at low concentrations. Among them, compounds 3b and 3p showed superior inhibitory potential compared to known commercial inhibitors. Molecular docking studies also revealed important interactions between these compounds and the target enzymes.

Synthesis, in vitro bio-evaluation, and molecular docking study of thiosemicarbazone-based isatin/bis-Schiff base hybrid analogues as effective cholinesterase inhibitors

Summary: Isatin/bis-Schiff bases analogues based on thiocarbohydrazone were synthesized, characterized, and tested for their inhibitory activity against acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE) enzymes. All analogues showed good inhibitory potentials, with analogue 1 being the most effective inhibitor. Structure-activity relationship (SAR) analysis revealed the importance of substitution effects on phenyl rings. Molecular docking studies provided insight into the binding interactions of the most active inhibitors with the active sites of AChE and BuChE.

Temporal changes in risk of cardiovascular events in people with newly diagnosed type 2 diabetes with and without cardiovascular disease

Summary: The study aimed to examine temporal changes in the risk of cardiovascular events in individuals with newly diagnosed type 2 diabetes with and without cardiovascular disease. The results showed that individuals with newly diagnosed diabetes had an increased risk of cardiovascular events, but the risk significantly decreased from 1997 to 2014, with a greater decline in those with diabetes compared to controls.

Design and synthesis of chromone-nitrogen mustard derivatives and evaluation of anti-breast cancer activity

Summary: In this study, a series of nitrogen mustard derivatives of chromone were designed and synthesized, among which one compound showed the most potent antiproliferative activity against breast tumor cells and exhibited certain selectivity between tumor cells and normal cells. This compound possibly induced G2/M phase arrest and apoptosis in tumor cells by generating intracellular ROS and activating DNA damage, leading to inhibition of tumor cell metastasis, invasion, and adhesion.

In silico and in vitro studies of thiosemicarbazone-indole hybrid compounds as potent α-glycosidase inhibitors

Summary: In this study, the alpha-glucosidase inhibitory activity of a novel series of thiosemicarbazone-indole hybrid compounds was evaluated, followed by in silico studies on the screened compounds. Molecular docking and dynamics simulations further confirmed the stability of the complexes between the screened compounds and alpha-glucosidase.

Development and exploration of novel substituted thiosemicarbazones as inhibitors of aldose reductase via in vitro analysis and computational study

Summary: The role of aldose reductase (ALR2) in causing diabetic complications is well-studied. In this study, a series of N-substituted thiosemicarbazones with added phenolic moieties were synthesized, and compound 3m showed strong and selective ALR2 inhibitory activity in vitro as well as promising antioxidant activity. Molecular docking studies and molecular dynamics simulations confirmed the stable interactions between 3m and ALR2. Therefore, N-substituted thiosemicarbazones are potential drug candidates for selective inhibition of ALR2 and treatment of diabetic complications.
