Recent advances in lipid nanoparticles for delivery of nucleic acid, mRNA, and gene editing-based therapeutics

Hidefumi Mukai et al.

Summary: Lipid nanoparticles (LNPs) are increasingly popular for delivering therapeutic drugs, especially in the field of nucleic acid and mRNA therapy. Future research will focus on expanding drug delivery scope, studying LNPs application in different human tissues, and investigating LNPs' pharmacokinetic changes under physiological and pathological conditions.

Focused ultrasound/microbubbles-assisted BBB opening enhances LNP-mediated mRNA delivery to brain

Koki Ogawa et al.

Summary: This study investigated the use of microbubble-assisted focused ultrasound blood-brain barrier opening for the intravenous delivery of mRNA-LNP. The results showed that exogenous protein expression by mRNA-LNP occurred only when FUS and microbubbles were applied to the brain.

Synthesis and evaluation of a novel adapter lipid derivative for preparation of cyclic peptide-modified PEGylated liposomes: Application of cyclic RGD peptide

Naoya Kato et al.

Summary: Peptide ligand modified nanoparticles can be easily prepared using the post-insertion method, which requires highly water dispersible peptide-lipid conjugates. In this study, a novel adapter lipid derivative was synthesized using the click reaction, enabling the synthesis of various cyclic peptides and the preparation of cRGD-modified PEGylated liposomes with good water dispersibility. The resulting liposomes efficiently associated with integrin αvβ3-expressing colon carcinoma cells and exhibited high cytotoxicity when loaded with doxorubicin.

Stability Study of mRNA-Lipid Nanoparticles Exposed to Various Conditions Based on the Evaluation between Physicochemical Properties and Their Relation with Protein Expression Ability

Mariko Kamiya et al.

Summary: This study evaluated the effects of temperature, cryoprotectants, vibration, light exposure, and syringe aspiration on the physicochemical properties of mRNA-LNPs. It was found that storage at -80 degrees C without a cryoprotectant caused a decrease in protein expression. Similar changes were observed with exposure to vibration and light under certain conditions.

RGD peptide-based lipids for targeted mRNA delivery and gene editing applications

Jingya Qin et al.

Summary: The designed RGD peptide based ionizable lipids for LNPs showed promising specific mRNA delivery both in vitro and in vivo, with increased expression in the whole body. This RGD-based hybrid LNP system holds potential for targeted mRNA delivery, allowing for more efficient and specific mRNA-based protein replacement and gene editing with reduced off-target effects.

The mRNA-LNP platform's lipid nanoparticle component used in preclinical vaccine studies is highly inflammatory

Sonia Ndeupen et al.

Summary: Vaccines based on mRNA-containing LNPs are a promising platform for COVID-19 vaccines, but may lead to highly inflammatory responses. Studies have shown that LNPs can induce massive neutrophil infiltration and activation of various inflammatory pathways in mice, which may be one of the factors affecting their immunogenicity and side effects.
