Dimethyl Fumarate Combined With Vemurafenib Enhances Anti-Melanoma Efficacy via Inhibiting the Hippo/YAP, NRF2-ARE, and AKT/mTOR/ERK Pathways in A375 Melanoma Cells

Hongxia Li et al.

Summary: Combination treatment of dimethyl fumarate (DMF) and vemurafenib (Vem) exhibits enhanced antitumor efficacy in melanoma by inhibiting multiple cell signaling pathways, leading to inhibition of cell proliferation and induction of cell death.

Muscle 4EBP1 activation modifies the structure and function of the neuromuscular junction in mice

Seok-Ting J. Ang et al.

Summary: Dysregulation of mTORC1 activity leads to NMJ structural instability during aging. This study investigates the roles of two mTORC1 phosphorylation targets, S6K1 and 4EBP1, in regulating NMJ structural instability caused by aging and sustained mTORC1 activation. The findings suggest that activation of 4EBP1 can induce NMJ remodeling and enhance synaptic transmission by activating satellite cells and increasing post-synaptic AChR turnover.

Mechanisms of muscle atrophy and hypertrophy: implications in health and disease

Roberta Sartori et al.

Summary: Skeletal muscle acts as a protein reservoir in the body and plays a key role in regulating glucose and lipid levels. The growth or loss of muscle mass can impact overall metabolism, movement, eating, and breathing. Various quality systems control muscle function, adapting cells to environmental and nutritional cues.

Vemurafenib provides a rapid and robust clinical response in pediatric Langerhans cell histiocytosis with the BRAF V600E mutation but does not eliminate low-level minimal residual disease per ddPCR using cell-free circulating DNA

Dmitry Evseev et al.

Summary: This study investigated the effectiveness of vemurafenib in children with BRAF V600E + LCH, demonstrating that the drug was effective in treating these children. However, it was found that vemurafenib did not completely eradicate the disease post-treatment, with some patients experiencing relapse after cessation of treatment. Furthermore, the long-term toxicity of vemurafenib has not been established and requires further research.

Molecular Mechanisms of Skeletal Muscle Hypertrophy

Stefano Schiaffino et al.

Summary: Skeletal muscle hypertrophy can be induced through various mechanisms, such as direct and indirect actions of hormones and growth factors, as well as the mediation of mechanical signals. Muscle growth is regulated at both the translational and transcriptional levels, with factors like mTORC1, transcription factors, and co-activators playing important roles in the process.
