Timing of maternal immune activation and sex influence schizophrenia-relevant cognitive constructs and neuregulin and GABAergic pathways

Maternal Immune Activation and Interleukin 17A in the Pathogenesis of Autistic Spectrum Disorder and Why It Matters in the COVID-19 Era

Michael Carter et al.

Summary: Autism spectrum disorder (ASD) is a common neurodevelopmental disability with a complex etiology involving genetic and environmental factors. Maternal immune activation (MIA) due to bacterial or viral infection during pregnancy has received significant attention as an environmental exposure that may influence neurodevelopmental outcomes in offspring. Interleukin-17A is suggested to mediate the effects of MIA on neurodevelopment. Monitoring neurodevelopmental outcomes in children exposed to MIA induced by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) during gestation is important.

Virus-Induced Maternal Immune Activation as an Environmental Factor in the Etiology of Autism and Schizophrenia

Aicha Massarali et al.

Summary: Maternal immune activation is a phenomenon that is caused by the activation of inflammatory pathways and affects fetal neural development. Maternal viral infection is one of the significant factors that lead to immune activation in pregnant women. Maternal immune activation and immune abnormalities play key roles in the development of conditions such as autism and schizophrenia. The methods of studying maternal immune activation include animal models and human epidemiological studies, but both have limitations.

Microglial GPR56 is the molecular target of maternal immune activation-induced parvalbumin-positive interneuron deficits

Diankun Yu et al.

Summary: Parvalbumin-positive (PV+) interneurons are vital for circuit rhythm maintenance and their decrease has been linked to autism spectrum disorders. Animal studies have shown that maternal immune activation (MIA) can lead to reduced PV+ interneurons and autism-like behaviors. This study reveals that MIA down-regulates microglial Gpr56 expression, and its deletion mimics MIA-induced interneuron defects and autism-like behaviors. Additionally, elevated microglial tumor necrosis factor-alpha expression is implicated in impairing interneuron generation. Genetically restoring Gpr56 expression in microglia can improve PV+ interneuron deficits and autism-like behaviors in offspring. Therefore, microglial GPR56 plays an important role in PV+ interneuron development and is a significant target in MIA-induced neurodevelopmental disorders.

Prenatal immune stress blunts microglia reactivity, impairing neurocircuitry

Sex-specific behavioral outcomes of early-life adversity and emerging microglia-dependent mechanisms

Madison M. Garvin et al.

Summary: Early-life adversity has significant effects on brain circuit maturation and vulnerability to cognitive and emotional disorders. The importance of considering sex as a biological variable in studying the effects of early-life adversity has only recently been recognized. This article discusses the sex-specific behavioral outcomes of early-life adversity in both humans and animal models and proposes microglia-mediated mechanisms as a potential underlying cause. Research in rodent models suggests that early-life adversity primarily affects males, leading to cognitive deficits, anhedonia, and alcohol abuse, while females exhibit different behavioral outcomes such as risk-taking and opioid addiction-related behaviors. Microglia have been identified as a key player in the effects of early-life adversity, with sex differences in synaptic rewiring and neural circuit development contributing to the divergent outcomes observed in males and females.

Microglia exhibit a robust proliferative capacity during development and in neurodegenerative diseases, but this capacity is largely absent at homeostasis. Proper microglial function in different states is heavily dependent on density fluctuations primarily induced by proliferation.
