Synthetic protocols for preparing these pyridopyrimidine derivatives are considered. The review is organized into four sections, categorized by the type of pyridopyrimidine. For each compound, the biological activity and synthetic route are presented. The bibliographic research for each type of pyridopyrimidine was conducted using Reaxys and Scifinder.

Synthesis of pyrido[2,3-d]pyrimidines can be from pyrimidines derivatives or pyridine derivatives. Various reported biological activities of pyrido[2,3-d]pyrimidines like selective adenosine kinase inhibitors, analgesic, anti-inflammatory, antimicrobial and herbicides are also discussed.

Synthesis and antitumor activity of novel pyridino[2,3-d]pyrimidine urea derivatives

Summary: A series of newly synthesized compounds exhibited significant inhibitory effects against MCF-7 and HCT-116 cells, with compounds 9g and 8b showing the most significant inhibitory effects. The synthesized pyridine[2,3-d]pyrimidine derivatives containing a urea group also demonstrated antitumor activities against MCF-7 and HCT-116 cells in vitro.

Synthesis and Biological Activity Studies of Novel Pyrido[2,3-d]pyrimidines and Pyrido[2,3-d]triazines

Summary: Novel derivatives of pyrido[2,3-d]pyrimidin-2,4(1H,3H)-dithione, 2,3-dimethylpyrido[2,3-d]pyrimidin-4-one, 4-chloro-2-methypyrido[2,3-d]pyrimidine, and pyrido[2,3-d]1,2,3-triazin-4-one demonstrated excellent anti-microbial activities. Most of the compounds also exhibited high safety profiles in cytotoxicity tests, with a few exceptions. Spectral data and elemental analysis confirmed the structures of the newly synthesized compounds.

Facile One-Pot Multicomponent Synthesis of Pyrazolo-Thiazole Substituted Pyridines with Potential Anti-Proliferative Activity: Synthesis, In Vitro and In Silico Studies

Summary: Pyrazolothiazole-substituted pyridine conjugates are a class of heterocyclic compounds with potential medicinal and pharmacological applications. A facile synthesis method for a new pyrazolo-thiazolo-pyridine conjugate 4 was described, which was utilized as a base for the synthesis of 15 novel pyrazolothiazole-based pyridine conjugates. In vitro evaluation of the synthesized compounds showed strong anti-proliferative activity, with compound 7 exhibiting the most potent cytotoxicity compared to etoposide. Additionally, molecular docking studies indicated that compound 7 has high binding affinity towards topoisomerase II, suggesting it as a promising lead anticancer compound.
