Chromatin alterations during the epididymal maturation of mouse sperm refine the paternally inherited epigenome

Yudhishtar S. Bedi et al.

Summary: Paternal lifestyle choices and exposure history can influence the health and fitness of the next generation. This study reveals that histone posttranslational modifications change during epididymal maturation, which may have implications for germline programming.

Alcohol induced increases in sperm Histone H3 lysine 4 trimethylation correlate with increased placental CTCF occupancy and altered developmental programming

Yudhishtar S. Bedi et al.

Summary: Using a mouse model, this study shows that paternal preconception alcohol intake affects offspring fetal-placental growth, with long-lasting consequences on adult metabolism. The researchers also find that chronic preconception male alcohol exposure leads to changes in sperm histone enrichment, which are associated with altered developmental programming in the placenta.

Recruitment of CTCF to an Fto enhancer is responsible for transgenerational inheritance of BPA-induced obesity

Yoon Hee Jung et al.

Summary: This study demonstrates that exposure of pregnant mice to bisphenol A (BPA) can lead to transgenerational transmission of obesity in F2 progeny up to the F6 generation. Chromatin accessibility analysis reveals alterations near binding motifs for CCCTC-binding factor (CTCF) at two cis-regulatory elements of the Fto gene, which correlate with the transmission of obesity. The interaction between these cis-regulatory elements and the Irx3 and Irx5 genes, involved in the differentiation of appetite-controlling neurons, is increased in sperm of obese mice. Deletion of the CTCF site in Fto results in mice that have normal food intake and fail to become obese when ancestrally exposed to BPA.

Loss of the cleaved-protamine 2 domain leads to incomplete histone-to-protamine exchange and infertility in mice

Lena Arevalo et al.

Summary: Protamines are essential sperm-specific proteins that package and protect paternal chromatin. The cP2 domain plays a crucial role in sperm chromatin condensation, and its deficiency leads to DNA fragmentation and infertility. This study provides insights into the function of the cP2 domain and opens up avenues for further research.

Paternally inherited H3K27me3 affects chromatin accessibility in mouse embryos produced by round spermatid injection

Mizuki Sakamoto et al.

Summary: Round spermatid injection (ROSI) results in a lower birth rate than intracytoplasmic sperm injection due to epigenetic abnormalities resulting in inefficient development of ROSI embryos. In this study, we found that a repressive histone mark, H3K27me3, persists in ROSI embryos and is associated with less accessible chromatin and impaired gene expression. The absence of epigenetic remodelling during spermiogenesis leads to dysregulation of chromatin accessibility and transcription in ROSI embryos, highlighting the importance of epigenetic remodelling in successful reproduction.

H4K5 Butyrylation Coexist with Acetylation during Human Spermiogenesis and Are Retained in the Mature Sperm Chromatin

Alberto de la Iglesia et al.

Summary: The study reveals the dual behavior of H4K5bu and H4K5ac in human spermatogenesis, with both modifications present at different stages of sperm development. These modifications remain stable under different testicular pathologies and are retained in ejaculated sperm. However, no direct association between these modifications and sperm quality was observed.

Protamines: lessons learned from mouse models

Lena Arevalo et al.

Summary: Protamines package and shield paternal DNA in the sperm nucleus, and their function and interactions with other sperm nuclear proteins show surprising complexity. This review provides an update on our understanding of protamines and their role in chromatin organization and male fertility using mouse models. Both PRM1 and PRM2 are essential for the production of functional sperm, with PRM1 playing a role in motility in addition to DNA compaction. Loss of one allele of PRM1 leads to subfertility, while heterozygous loss of PRM2 does not have a significant impact. Unprocessed PRM2 seems to be involved in the eviction of intermediate DNA-bound proteins and the incorporation of protamines into chromatin.

Dynamic nucleosome organization after fertilization reveals regulatory factors for mouse zygotic genome activation

Chenfei Wang et al.

Summary: This study investigated the dynamic changes in nucleosome occupancy and positioning in pronucleus-stage zygotes using a new method. The researchers found that nucleosomes were established more quickly in the paternal genome and were associated with specific transcription factor binding sites. These findings have important implications for understanding the mechanisms of chromatin remodeling after fertilization and key transcription regulators during development.

Histone H3 lysine 4 trimethylation in sperm is transmitted to the embryo and associated with diet-induced phenotypes in the offspring

Mechanisms of maternal intergenerational epigenetic inheritance

Andrina Staubli et al.

Summary: In this study, the major dynamic changes in transcriptional processes, chromatin composition, and 3D organization of maternal and paternal genomes during oocyte and early embryonic development in mice were summarized. The study provides mechanistic insights into molecular hierarchies and crosstalk between various chromatin-associated processes, defining three distinct types of maternal epigenetic memory states in oocytes to support pre-implantation and post-implantation development.

Widespread displacement of DNA- and RNA-binding factors underlies toxicity of arginine-rich cell-penetrating peptides

Vanesa Lafarga et al.

Summary: The presence of arginine-rich cell-penetrating peptides (CPPs) leads to displacement of RNA- and DNA-binding proteins from chromatin and mRNA, affecting nucleic acid-related reactions, potentially explaining the toxicity of these CPPs. The effects of (PR)n dipeptide repeat were found to be similar to those of protamine, a small arginine-rich protein that displaces histones during spermatogenesis.

H4K20me3 marks distal intergenic and repetitive regions in human mature spermatozoa

Nihan Ozturk et al.

Summary: The genome-wide distribution of the heterochromatic marker H4K20me3 was analyzed in human sperm and somatic cells, revealing similar patterns of enrichment in distal intergenic regions and repetitive gene clusters. H4K20me3-free nucleosomes were more common in genic regions and associated with genes encoding developmental factors. The evidence suggests that paternally transmitted histones may serve a dual purpose of maintaining heterochromatin and guiding transcription of euchromatin.

Whole-genome sequencing of H3K4me3 and DNA methylation in human sperm reveals regions of overlap linked to fertility and development

Romain Lambrot et al.

Summary: The study reveals the presence of H3K4me3 in sperm, which is distributed throughout the genome and found at genes related to fertility and development. Additionally, enrichment of H3K4me3 is observed at regions that escape epigenetic reprogramming and overlap with DNA methylation, suggesting a potential interplay between these marks in sperm. Comparisons with the embryonic transcriptome indicate an influence of paternal chromatin on embryonic gene expression.

Reprogramming of the histone H3.3 landscape in the early mouse embryo

Takashi Ishiuchi et al.

Summary: Epigenetic reprogramming of the zygote involves the dynamic incorporation of histone variant H3.3, with a non-canonical pattern present in mature oocytes and zygotes transitioning to a canonical pattern at the two-cell stage. This transition is facilitated by the chromatin assembly factor CAF-1 mediating the incorporation of H3.1/H3.2.
