Integrated metabolomics and transcriptomics reveal the neuroprotective effect of nervonic acid on LPS-induced AD model mice

Xueqi Wang et al.

Summary: In this study, the neuroprotective effect of nervonic acid (NA) and its molecular mechanism in AD model mice were investigated using behavioral, transcriptomic, and metabolomic approaches. The results showed that NA improved motor skills and learning and memory abilities in mice. Metabolomic and transcriptomic analyses revealed that NA reversed the neuroinflammation induced by LPS and regulated multiple gene transcription and metabolism pathways. This study provides preliminary evidence for the molecular mechanism of NA.

White matter myelination during early infancy is linked to spatial gradients and myelin content at birth

Mareike Grotheer et al.

Summary: In this study, the development of white matter in infants from 0 to 6 months was investigated using diffusion MRI and quantitative MRI measurements. The researchers found that the longitudinal relaxation rate (R1) of white matter increased over time, with faster development in less mature areas of white matter in newborns.

Bundle myelin fraction (BMF) mapping of different white matter connections using microstructure informed tractography

Simona Schiavi et al.

Summary: This study introduces a new method called myelin streamline decomposition (MySD), which extends convex optimization modeling to microstructure informed tractography (COMMIT), allowing the deconvolution of the actual value measured by a microstructural map on each individual streamline to recover unique bundle specific myelin fractions (BMFs).

APOE4 impairs myelination via cholesterol dysregulation in oligodendrocytes

Joel W. Blanchard et al.

Summary: APOE4 has widespread effects on gene expression in the human brain, particularly in relation to cholesterol homeostasis and transport. Aberrant cholesterol deposition in oligodendrocytes leads to reduced myelination, while pharmacologically facilitating cholesterol transport improves learning and memory. This study provides insights into the impact of APOE4 on the aging human brain and suggests potential therapeutic strategies for Alzheimer's disease.

The role of very long chain fatty acids in yeast physiology and human diseases

Pia Erdbruegger et al.

Summary: VLCFAs are highly diverse molecules with variable chain length, double bond numbers, and hydroxylation sites, synthesized in the ER through membrane embedded enzymes. Mis-regulation of VLCFA metabolism can lead to inherited diseases like ichthyosis and demyelination. Recent studies in yeast suggest intricate regulation of VLCFA metabolism for lipid homeostasis.

Ceramide synthase 2-C24:1-ceramide axis limits the metastatic potential of ovarian cancer cells

Xuewei Zhang et al.

Summary: Regulation of sphingolipid metabolism has a significant impact on cellular homeostasis and cancer progression. Dysregulation of ceramide-centered metabolism is associated with metastatic potential in ovarian cancer cells. Specifically, the CerS2-C-24:1-ceramide axis, possibly counteracted by neutral ceramidase, plays a key role in limiting cell motility and metastatic potential.

Individual non-esterified fatty acids and incident atrial fibrillation late in life

Cara N. Pellegrini et al.

Summary: In this study, higher serum levels of non-esterified nervonic acid were positively associated, while higher levels of non-esterified gamma-linolenic acid were inversely associated with incident atrial fibrillation (AF) among older adults. These results may provide new strategies for AF prevention and early intervention in this high-risk population segment.

Plasmalogens and Chronic Inflammatory Diseases

Jose Carlos Bozelli et al.

Summary: Lipids play key roles in cellular function, with plasmalogens receiving attention due to their association with degenerative and metabolic disorders. Inflammatory processes can affect plasmalogen levels and trigger either anti- or pro-inflammatory responses. Restoring plasmalogen levels has been shown to be a successful anti-inflammatory strategy.
