Image-based deep learning identifies glioblastoma risk groups with genomic and transcriptomic heterogeneity: a multi-center study

Jing Yan et al.

Summary: This study developed a deep learning imaging signature (DLIS) for risk stratification in patients with multiforme (GBM) and investigated the biological pathways and genetic alterations underlying DLIS. DLIS was associated with survival and an independent predictor. The integrated nomogram incorporating DLIS achieved improved prediction performance compared to the clinicomolecular nomogram. DLIS was correlated with core pathways of GBM and key genetic alterations.

The survival outcomes of molecular glioblastoma IDH-wildtype: a multicenter study

Andres Ramos-Fresnedo et al.

Summary: This study aimed to compare the survival outcomes between molecular GBM (molGBM) and histological GBM (histGBM). The results showed that molGBM had a longer progression-free survival (PFS) compared to histGBM, but similar overall survival (OS).

A novel patient stratification strategy to enhance the therapeutic efficacy of dasatinib in glioblastoma

Obada T. Alhalabi et al.

Summary: The mesenchymal subtype of glioblastoma showed higher sensitivity to dasatinib due to increased SRC pathway activation, with SERPINH1 suggesting a potential predictive marker. Stratification based on gene expression subtyping should be considered in future dasatinib trials for better patient selection strategies.

Synaptic input to brain tumors: clinical implication

Varun Venkataramani et al.

Summary: The discovery of synaptic connections between neurons and brain tumor cells challenges current understandings of gliomas and brain metastases, showing how these tumors integrate into complex neuronal circuits. The study provides insights into neuronal-brain tumor synaptic communication (NBTSC) and potential therapeutic avenues, highlighting promising future targets to reveal critical vulnerabilities of incurable brain tumors.

Summary: Radiomic analysis provides a powerful tool for extracting clinically relevant information from radiologic imaging, but the data-driven nature of radiomics inherently lacks insights into the biological underpinnings of observed relationships.

Deep learning features from diffusion tensor imaging improve glioma stratification and identify risk groups with distinct molecular pathway activities

Jing Yan et al.

Summary: A deep learning signature (DLS) from diffusion tensor imaging (DTI) was developed for predicting overall survival in patients with infiltrative gliomas, with five key biological pathways identified. The DLS was associated with survival, independent predictor, and improved survival prediction when incorporated into existing risk system. Therapies targeting neuron-to-brain tumor synaptic communication may be more effective in high-risk glioma defined by DTI-derived DLS.

Biologic Pathways Underlying Prognostic Radiomics Phenotypes from Paired MRI and RNA Sequencing in Glioblastoma

Qiuchang Sun et al.

Summary: This study investigated the biological significance of radiomics in glioblastomas using four datasets. By analyzing MRI and RNA sequencing data, four prognostic radiomics phenotypes correlated with distinct biological pathways were identified, and the importance of the RadGene score in prognosis was confirmed.

Quantitative MRI-based radiomics for noninvasively predicting molecular subtypes and survival in glioma patients

Jing Yan et al.

Summary: MRI-based radiomics can be used for noninvasively detecting molecular groups and predicting survival in gliomas, regardless of grades. The image fusion model achieved high accuracy in predicting IDH and TERT status, while the cT1WI-based radiomic signature alone performed well in predicting 1p/19q status. Predictive molecular groups showed comparable prognostic value to actual ones in predicting PFS and OS. Prognostic nomograms based on grades and predictive molecular groups yielded good predictive performance for PFS and OS.
