Does Food Affect the Pharmacokinetics of Non-orally Delivered Drugs? A Review of Currently Available Evidence

Peng Zou

Summary: The food effects on orally administered drugs have been extensively studied, but there is limited knowledge about the food effects for non-orally administered drugs. This review paper collected clinical data on food effects for non-orally administered drugs through a literature survey. The findings suggest that food intake can affect the absorption and elimination of different types of non-orally administered drugs, with potential implications for efficacy and safety.

Short- and long-term effects of body weight loss following calorie restriction and gastric bypass on CYP3A-activity - a non-randomized three-armed controlled trial

Kine Eide Kvitne et al.

Summary: This study compared the effects of RYGB and calorie restriction on CYP3A activity, finding that neither RYGB surgery nor weight loss impacted the absolute bioavailability or clearance of midazolam in the short term. In the long term, both RYGB and diet groups showed a decrease in absolute bioavailability of midazolam, suggesting that CYP3A activity recovery is not solely dependent on weight loss through RYGB.

Determination of CYP450 Expression Levels in the Human Small Intestine by Mass Spectrometry-Based Targeted Proteomics

Alexia Grangeon et al.

Summary: This study quantified the protein expression levels of 16 major CYP450 isoforms in various sections of the human small intestine using LC-MS/MS, revealing seven detected isoforms and observing wide inter-subject variability. The study found that CYP450 protein levels were better correlated with specific isoform activities than with mRNA levels, which could provide new data for improving physiologically based pharmacokinetic models.

Influence of Inflammation on Cytochromes P450 Activity in Adults: A Systematic Review of the Literature

Camille Lenoir et al.

Summary: Inflammation has a specific impact on CYP activities, depending on the nature and severity of the underlying inflammatory disease. The drug-disease interactions can significantly affect drug pharmacokinetic parameters and clinical management. Resolution of inflammation may lead to normalization of CYP activity, but further research is needed to clarify some equivocal results.
