Late-onset enteric virus infection associated with hepatitis (EVAH) in transplanted SCID patients

Quentin Riller et al.

Summary: Persistent hepatitis is a common late-onset posttreatment complication in transplanted SCID patients. This study found that it is associated with chronic enteric viral infection and immune dysregulation, and identified risk factors such as myeloablation absence and defective B-cell function. Treatment options such as retransplantation or gene therapy can lead to remission of hepatitis.

Immune dysregulation

Andrew Long et al.

Summary: The understanding of immune dysregulation in various diseases continues to expand, with a focus on the role of altered microbiome, gut barrier dysfunction, and age-associated B cells in systemic inflammation. T follicular regulatory cells and their potential therapeutic interventions are also highlighted. Studies on juvenile idiopathic arthritis and hyperinflammatory syndromes provide insights into their pathogenesis and potential biomarkers and therapeutic targets for these conditions. Future trials are needed to address remaining questions regarding the pathogenesis, diagnosis, and treatment of autoimmune, inflammatory, and immunodeficiency syndromes.

Human Inborn Errors of Immunity: 2022 Update on the Classification from the International Union of Immunological Societies Expert Committee

Stuart G. Tangye et al.

Summary: The International Union of Immunological Societies Expert Committee has published an updated classification of inborn errors of immunity, which includes 55 novel monogenic gene defects and 1 phenocopy caused by autoantibodies. This update contributes to our understanding of the molecular, cellular, and immunological mechanisms of human immune diseases, and provides a resource for molecular diagnosis of heritable immunological disorders and scientific research on monogenic and related human immune diseases.

Microseek: A Protein-Based Metagenomic Pipeline for Virus Diagnostic and Discovery

Philippe Perot et al.

Summary: Microseek is a pipeline for virus identification and discovery based on the RVDB-prot database. It analyzes mNGS raw data and determines viral sequences through Lowest Common Ancestor scoring. Experimental results on human samples demonstrate that Microseek performs well in identifying known and distant pseudoviral sequences while minimizing non-relevant results.

Identification of Umbre Orthobunyavirus as a Novel Zoonotic Virus Responsible for Lethal Encephalitis in 2 French Patients with Hypogammaglobulinemia

Philippe Perot et al.

Summary: The study identified Umbre arbovirus in the brain tissue of two immunocompromised patients, representing a previously unidentified orthobunyavirus in humans. In addition, viral sequences related to the virus were found in mosquitoes in southern France, indicating the presence of similar arboviruses in Europe and their potential public health impact.

Interstitial Lung Disease in Common Variable Immunodeficiency

Joao Pedro Lopes et al.

Summary: Interstitial lung disease (ILD) is a common complication in patients with common variable immunodeficiency (CVID), often associated with bronchiectasis and autoimmunity. Lung biopsy pathology in CVID patients with ILD commonly shows granulomas and lymphoid interstitial pneumonia. Autoimmune manifestations are common, and ILD in CVID can lead to significant mortality, highlighting the need for further research on optimal management strategies.
