A novel homozygous mutation in LSS gene possibly causes hypotrichosis simplex in two siblings of a Tibetan family from the western Sichuan province of China

Bei Zhao et al.

Summary: This study aims to find mutations in the LSS gene from a Chinese family affected by hypotrichosis simplex. The results show that the affected individuals have a new homozygous mutation in the LSS gene, while their asymptomatic parents have a heterozygous mutation. The mutation may lead to reduced enzyme activity and contribute to the development of hypotrichosis simplex.

Summary: This study investigates the contribution of LSS mutations to mutilating palmoplantar keratoderma (PPK) in a Chinese patient. Biallelic variants were identified in the LSS gene, and experiments showed a correlation between these mutations and the disease.

Biallelic Variants in Lanosterol Synthase (LSS) Cause Palmoplantar Keratoderma- Congenital Alopecia Syndrome Type 2

Fang Yang et al.

Summary: This study identified LSS gene variants associated with palmoplantar keratoderma-congenital alopecia syndrome type 2, resulting in loss of enzymatic activity and abnormal cornified envelope formation. These findings highlight the importance of the cholesterol synthesis pathway in human skin cornification.

Expanding the Phenotypic Spectrum of APMR4 Syndrome Caused by a Novel Variant in LSS Gene and Review of Literature

Nesma M. Elaraby et al.

Summary: Alopecia intellectual disability syndromes 4 (APMR4) is a rare genetic condition caused by a mutation in the LSS gene, leading to intellectual disability and congenital alopecia. This study reports a new pathogenic variant discovered through whole-exome sequencing in an Egyptian family with affected siblings and a prenatally diagnosed fetus. The findings expand the understanding of the syndrome's clinical and mutational spectrum, emphasizing the importance of prenatal testing and genetic counseling.

Summary: This study aimed to definitively diagnose three pediatric patients suspected of HS in two unrelated Chinese families through whole-exome sequencing, identifying novel accompanying phenotypes such as teeth dysplasia, and inferring potential genotype-phenotype correlations for LSS mutations for the first time.

Alopecia-mental retardation syndrome: Molecular genetics of a rare neuro-dermal disorder

Muhammad Muzammal et al.

Summary: APMR syndrome is a rare autosomal recessive neuro-dermal disorder characterized by absence of hair on the scalp, eyelashes, and eyebrows, along with intellectual disability. Approximately 14 families have been reported with APMR globally, with a prevalence of 1 in 1,000,000 persons.

Summary: Bi-allelic mutations in the lanosterol synthase (LSS) gene have been shown to underlie both non-syndromic and syndromic forms of hypotrichosis. This study identified two Japanese patients with severe congenital hypotrichosis carrying bi-allelic mutations in the LSS gene. The findings emphasize the essential roles of the LSS gene in hair follicle development and hair growth in humans.
