Summary: Sepsis is a leading cause of illness and death worldwide, and its associated coagulation disorders significantly worsen the prognosis. During the COVID-19 pandemic, research has focused on the pathogenesis and treatment of SARS-CoV-2. Coagulation disturbances have been found to contribute to severe outcomes of SARS-CoV-2 infection. Similarities between COVID-19-associated coagulopathy and sepsis-induced coagulopathy have been observed, but their relationship is yet to be established. Understanding the pathogenesis of COVID-19-associated coagulopathy is crucial for accurate definitions and classification criteria.

Monocyte distribution width alterations and cytokine storm are modulated by circulating histones

Summary: The study investigated the role of extracellular histones on monocyte distribution width (MDW) and their effect on the release of cytokines by blood cells. The results showed that extracellular histones can induce changes in MDW and promote heterogeneity in monocytes. They also lead to the release of cytokines, especially G-CSF, IL-1 beta, IL-6, MIP-1 beta, and IL-8. These findings may have significant implications for predicting the risk of severe outcomes.

Circulating histones contribute to monocyte and MDW alterations as common mediators in classical and COVID-19 sepsis

Summary: This study found that histones can significantly increase the monocyte distribution width (MDW) index in whole blood samples from healthy individuals when treated in vitro, similar to the changes found in patients with classic and viral sepsis. These changes are associated with histone-induced modifications of monocyte volume, cytoplasmic granularity, vacuolization, and nuclear structure. Therefore, histones may play a crucial role in the observed alterations of monocytes in acute classical and viral sepsis.
