Fatty acid-binding protein 5 aggravates pulmonary artery fibrosis in pulmonary hypertension secondary to left heart disease via activating wnt/β-catenin pathway

Qian Lei et al.

Summary: This study reveals the significant role of fatty acid-binding protein 5 (FABP5) in pulmonary hypertension secondary to left heart disease (PH-LHD). Through experiments and in vitro studies, it was demonstrated that inhibiting FABP5 can improve pulmonary vascular remodeling and cardiac function. FABP5 may serve as a potential therapeutic target for PH-LHD.

Inhibition of PIKfyve Ameliorates the Proliferation and Migration of Vascular Smooth Muscle Cells and Vascular Intima Hyperplasia By Reducing mTORC1 Activity

Min Gu et al.

Summary: This study investigated the role and mechanism of PIKfyve in the proliferation and migration of vascular smooth muscle cells (VSMCs) and vascular intima hyperplasia. The results showed that inhibiting PIKfyve could ameliorate the enhanced activity of VSMC proliferation and migration induced by platelet-derived growth factor (PDGF)-BB. The study suggests that targeting PIKfyve might be a novel effective method to reduce the proliferation and migration of VSMCs and vascular restenosis.

P2X7 Receptors: An Untapped Target for the Management of Cardiovascular Disease

Brandon G. Shokoples et al.

Summary: Chronic low-grade inflammation is closely linked to cardiovascular diseases. Targeting the purinergic receptor P2X7, which mediates inflammation and cellular death, shows potential in mitigating cardiovascular conditions. Inhibiting P2X7 activation and IL-1 beta/IL-18 release may reduce cardiovascular disease severity and occurrence of adverse cardiac events.

Danlou Tablet Activates Autophagy of Vascular Adventitial Fibroblasts Through PI3K/Akt/mTOR to Protect Cells From Damage Caused by Atherosclerosis

Li Wang et al.

Summary: Danlou tablet (DLT) activates autophagy in vascular adventitial fibroblasts to protect cells from atherosclerosis damage. DLT inhibits lipid accumulation and fibrosis formation, reduces serum lipid levels, inflammatory factors, and suppresses PI3K/Akt/mTOR pathway to achieve this purpose.

P2X7 Receptor-Mediated Inflammation in Cardiovascular Disease

Junteng Zhou et al.

Summary: Activation of the P2X7 receptor mediates inflammatory responses and pyroptosis in cardiovascular diseases, leading to the release of pro-inflammatory cytokines and worsening cardiac phenotypes. Inhibition of the P2X7 receptor has shown promise in attenuating these pathological mechanisms and improving cardiovascular outcomes.

Loss of the transcriptional repressor Rev-erbα upregulates metabolism and proliferation in cultured mouse embryonic fibroblasts

Sean P. Gillis et al.

Summary: Loss of Rev-erb alpha in MEFs leads to a hypermetabolic phenotype with increased glycolysis and oxidative phosphorylation. Rev-erb alpha deletion upregulates genes related to glycolysis and the PPP, impacting cell function and proliferation. Knockout of Rev-erb alpha reduces cell viability and antioxidant defense capacity, revealing novel mechanisms by which Rev-erb alpha modulates glycolysis and the PPP.

SFRP5 inhibits melanin synthesis of melanocytes in vitiligo by suppressing the Wnt/β-catenin signaling

Dao-Pei Zou et al.

Summary: SFRP5 overexpression inhibits melanin synthesis and pigmentation in vitiligo by downregulating MITF and the Wnt signaling pathway, while SFRP5 silencing increases melanin synthesis and activates Wnt signaling. Additionally, SFRP5 overexpression downregulates TCF/LEF activity in melanocytes and the inhibitory effect on melanin synthesis can be reversed by a beta-catenin agonist.
