Immunodominant conformational and linear IgE epitopes lie in a single segment of Ara h 2

Summary: This study investigated the distribution of conformational epitopes on peanut allergens Ara h 2 and Ara h 6. Chimeric 2S-albumins were generated and their epitope specificity was determined using competitive inhibition assays. The results showed that the configuration of conformational epitopes was preserved in the chimeras and identified a critical segment for the IgE-binding epitope of Ara h 2. These findings provide new insights into the allergenic potency of peanut allergens.

Summary: This study aims to determine the mechanism underlying the unique susceptibility to anaphylaxis in C3H/HeJ mice and test the impact of Tlr4 and Dock8 mutations. The results showed increased gut permeability and an increased number of goblet cells in the small intestine of C3H/HeJ mice compared to C57BL/6 mice, leading to the entry of ingested allergens into the bloodstream and triggering anaphylactic reactions.
