Changing characteristics, treatment approaches and survival of patients with brain metastasis: data from six thousand and thirty-one individuals over an observation period of 30 years

Ariane Steindl et al.

Summary: Patient classification is crucial for guiding individualized treatment decisions and forming brain metastases (BMs) cohorts for clinical trials. The study reveals shifting trends in the presentation and treatment strategies of BM patients over the past decades. The presence of targetable driver mutations affects the clinical presentation and prognosis, emphasizing the need to address the modern composition of BM cohorts and the distinct clinical course of patients with targetable driver mutations in future BM-specific trials.

Bone metastases from differentiated thyroid carcinoma: heterogenous tumor response to radioactive Iodine therapy and overall survival

Arnaud Jannin et al.

Summary: This study aimed to evaluate the overall survival and prognostic factors of differentiated thyroid carcinoma patients with bone metastases, as well as the effectiveness of radioiodine treatment. The study found that approximately one-third of patients achieved a complete bone metastasis response following radioiodine treatment. The presence of (18)FDG uptake in bone metastases was associated with poor overall survival.

Molecular diagnosis and targeted treatment of advanced follicular cell-derived thyroid cancer in the precision medicine era

Jaume Capdevila et al.

Summary: Most malignant thyroid tumours are initially treated with surgery or a combination of surgery and radioactive iodine therapy. However, in patients with metastatic disease, many tumours become refractory to radioactive iodine (RAI), and these patients require alternative treatments, such as locoregional therapies and/or systemic treatment with multikinase inhibitors. Targeted therapies based on genetic alterations have shown clinical efficacy in thyroid cancer, including NTRK gene fusions, RET and BRAF inhibitions. Effective testing strategies for these genetic alterations are crucial for identifying patients who could benefit from targeted treatments.
