Adipose Tissue Fibrosis in Obesity: Etiology and Challenges

Genevieve Marcelin et al.

Summary: Obesity is a chronic and progressive condition that affects whole-body energy balance and is linked to the development of comorbidities. Fibrotic adipose tissue caused by obesity can lead to metabolic dysfunctions such as insulin resistance. Targeting inflammation or fibrosis deposition may help improve insulin resistance and glucose homeostasis in individuals with obesity.

Adipose-tissue plasticity in health and disease

Alexander Sakers et al.

Summary: Adipose tissue, known as fat, plays a crucial role in regulating various physiological aspects of the body. Its high plasticity allows for significant changes in metabolism, structure, and phenotype in response to physiological stimuli. Limitations in this plasticity contribute to decreased or aberrant responses to physiological cues, leading to the progression of cardiometabolic disease.

Targeting p21Cip1 highly expressing cells in adipose tissue alleviates insulin resistance in obesity

Lichao Wang et al.

Summary: This study identifies a small population of p21 high-expressing cells that accumulate in adipose tissue with obesity and shows that intermittent clearance of these cells can prevent and alleviate insulin resistance. Inactivation of the NF-kB pathway within these cells attenuates insulin resistance. Additionally, a drug combination of dasatinib and quercetin can eliminate p21 high-expressing cells in human fat and mitigate insulin resistance.

Apolipoprotein A5, a unique modulator of fasting and postprandial triglycerides

Linda May-Zhang et al.

Summary: The discovery of APOA5 has raised questions about its role in lipid transport and TG homeostasis. APOA5 has been identified as a regulator of plasma TG levels and may also affect postprandial TG metabolism through influencing chylomicron formation and transport. Mysteries remain on how this liver-derived protein modulates TG homeostasis, especially via the gut.

Summary: Excess and dysfunctional adipose tissue plays an important role in metabolic diseases. This study describes a novel ceiling culture-based model of human mature white adipocytes that can transdifferentiate into beige adipocytes. The use of rosiglitazone and rapamycin can modulate transdifferentiation and enhance the expression of beige adipocyte-specific genes.

Apolipoprotein A-II, a Player in Multiple Processes and Diseases

Gabriela Florea et al.

Summary: Apolipoprotein A-II (apoA-II) plays an important role in lipid metabolism and may have different effects in humans and mice. It influences lipid metabolism and may be associated with vascular diseases. Findings regarding its role in atherogenesis have been conflicting. In humans, apoA-II deficiency has little effect on lipoprotein levels, while in mice it causes HDL deficit. Increased plasma apoA-II concentration leads to hypertriglyceridemia and lowers HDL levels, potentially causing type 2 diabetes.

Summary: This study investigated the structure and lipid distribution of LNPs in the presence of ApoE using small-angle neutron scattering. The binding of ApoE was found to cause a redistribution of lipids at the shell and core of LNPs, impacting their internal structure and mRNA release. The rearrangement of LNP components upon ApoE incubation may have implications for LNP endosomal escape.

Adipose tissue and insulin resistance in obese

Bulbul Ahmed et al.

Summary: This review discusses the potential mechanisms behind obesity-associated insulin resistance, such as dysfunctional adipose tissue, excessive release of free fatty acids, and the impact of lipotoxicity on cellular dysfunction leading to systemic inflammation and disruption of the insulin signaling pathway. It highlights that hepatic insulin resistance is the primary event that triggers peripheral tissue insulin resistance.
