Target sequencing reveals the association between variants in VAX1 and NSCL/P in Chinese population

Yue You et al.

Summary: This study identified 6 SNPs in the VAX1 gene that are associated with NSCL/P in the Western Han Chinese population. Five of these SNPs were predicted to affect transcription factor binding ability and were expression quantitative trait loci (eQTLs) of nearby genes in multiple tissues.

Genetic and epigenetic studies in non-syndromic oral clefts

Azeez Alade et al.

Summary: The etiology of non-syndromic oral clefts is complex and involves genetics, genomics, epigenetics, and stochastic factors. Various approaches have been used to understand the etiology, including linkage analysis, candidate gene association studies, genome-wide association studies, whole-genome sequencing, copy number variations, and epigenetics. This review shares the methods, genes, and loci reported in some studies.

Genetic models and approaches to study orofacial clefts

Elizabeth J. Leslie

Summary: This review describes the evidence for a genetic etiology in orofacial clefts (OFCs), statistical genetic approaches employed to identify genetic causes, and how the results have shaped our current understanding of the genetic architectures of syndromic and nonsyndromic OFCs.

Genetic markers for non-syndromic orofacial clefts in populations of European ancestry: a meta-analysis

Lara Slavec et al.

Summary: This study systematically reviewed the relevant genetic risk factors for non-syndromic orofacial cleft (nsOFC) in populations of European ancestry. The meta-analysis identified several genetic variants in genes/loci such as IRF6, GRHL3, 8q24, VAX1, TGFA, FOXE1, ABCA4, NOG, GREM1, AXIN2, DVL2, WNT3A, and WNT5A that are strongly associated with nsOFC phenotypes. These findings suggest that these genes have high potential as biomarkers for nsOFC in European populations.

Evaluation of genome-wide association signals for nonsyndromic cleft lip with or without cleft palate in a multiethnic Brazilian population

Renato Assis Machado et al.

Summary: This study provides evidence for the association of rs34246903 (4p16.2) with non-syndromic cleft lip with or without cleft palate (NSCL +/- P), as well as for the association of rs13317 within FGFR1 with NSCL +/- P, in a multiethnic Brazilian population.

Association of genetic polymorphisms of VAX1, MAFB, and NTN1 with nonsyndromic cleft lip with or without cleft palate in Chinese population

Li Peng et al.

Summary: This case-control study confirmed the association between ten single-nucleotide polymorphisms (SNPs) in six genes and nonsyndromic cleft lip with or without palate (NSCL/P) in the Chinese population. Specifically, VAX1 rs7078160, MAFB rs11696257, and NTN1 rs4791774 were found to be associated with NSCL/P.

Revisiting the embryogenesis of lip and palate development

Nigel L. Hammond et al.

Summary: Clefts of the lip and palate (CLP) are a major cause of congenital facial malformation globally, resulting from failure of fusion of the facial processes during embryogenesis. The high prevalence and healthcare burden imposed by CLP highlight the importance of understanding the molecular mechanisms driving facial development.

Role and mechanism of BMP4 in bone, craniofacial, and tooth development

Yuer Ye et al.

Summary: BMP4 plays a crucial role in craniofacial and skeletal development, particularly in tooth development. Mutations or deletions of this protein can lead to various tissue development defects. Therefore, studying BMP4 is important for understanding the development of bones, craniofacies, and teeth.

Summary: The study found that the rs7078160 polymorphism is significantly associated with non-syndromic cleft lip, with or without cleft palate. In the Han population in southern China, individuals with AG+AA genotypes have a higher risk of cleft lip compared to those with GG genotype.
