Fibrosis, atrial fibrillation and stroke: clinical updates and emerging mechanistic models

Summary: Currently, stroke risk assessment and mitigation in patients with atrial fibrillation are focused on clinical risk factors, with atrial fibrosis emerging as a unifying mechanism for both arrhythmia and thrombus formation. In patients with embolic stroke of undetermined source (ESUS), fibrosis is increasingly recognized as a major risk factor, although the underlying mechanism of this correlation remains unclear. Computational models are promising in identifying therapeutic targets and improving understanding of thrombus formation and arrhythmia induction in these patients.

Summary: Cellular senescence is a growth arrest state caused by various stresses that plays a role in tumor suppression, embryonic development, and wound healing. Senescence can impact the pathology of diseases by secreting inflammatory chemokines and immune modulators, ultimately leading to inflammation, tissue fibrosis, immunosenescence, and aging-related diseases such as atrial fibrillation. Targeting senescence and its secretory phenotype may potentially attenuate atrial fibrillation.
