Split light up aptamers as a probing tool for nucleic acids

Summary: Aptamers that enhance non-fluorescent dyes' fluorescence can be transformed into fluorescent sensors. The split light-up aptameric sensors (SLAS) are composed of two RNA or DNA strands and a fluorogenic organic dye, allowing for cost-efficient label-free nucleic acid detection with improved selectivity and simplified assay optimization. RNA-based SLAS can be utilized for intracellular monitoring and imaging of biological molecules.

Boronic Acid-Mediated Activity Control of Split 10-23 DNAzymes

Summary: The 10-23 DNAzyme is a Mg2+-dependent catalytic oligonucleotide that can cleave RNA in a sequence-specific manner. In this study, new split 10-23 DNAzymes were designed using nonfunctional fragments that, when assembled with Mg2+ ions, form boronate internucleoside linkages to restore DNAzyme activity. The best-performing system was identified through systematic evaluation, highlighting key features for controlling DNAzyme activity via boronate linkages.

Towards Higher Complexity in the RNA World: Hairpin Ribozyme Supported RNA Recombination

Summary: In the RNA world, exchange of sequence patches between two RNAs can lead to the generation of new RNA molecules with novel functionality through RNA supported RNA recombination. Research demonstrates that a designed HPR variant can cleave two different RNA molecules and subsequently recombine and ligate the resulting fragments, yielding functional recombination products with up to 34% efficiency.
