Organocatalytic stereoselective cyanosilylation of small ketones

Summary: Enzymatic stereoselectivity is often better than most chemical catalysts in the conversion of small substrates. A recent study has reported the development of broadly applicable confined organocatalysts for the highly enantioselective cyanosilylation reaction. The selectivity (98:2 enantiomeric ratio) of the catalyst towards the pharmaceutically relevant product exceeds any other catalyst class, including engineered biocatalysts.

Novel TYK2 Inhibitors with an N-(Methyl-d3)pyridazine-3-carboxamide Skeleton for the Treatment of Autoimmune Diseases

Summary: In this study, a series of novel derivatives that bind to the TYK2 pseudokinase domain were designed, synthesized, and evaluated. Compound 30 showed excellent inhibitory potency against STAT3 phosphorylation, along with good JAK isoform selectivity, in vivo and in vitro pharmacokinetic properties, and high oral effectiveness in autoimmune disease models. These findings suggest that compound 30 is a promising candidate for therapeutic applications in autoimmune diseases.

Iridium-catalyzed α-selective deuteration of alcohols

Summary: The development of chemoselective C(sp(3))-H deuteration is of great interest. In this study, the alpha-selective, iridium(iii)-bipyridonate-catalyzed hydrogen(H)/deuterium(D) isotope exchange of alcohols using deuterium oxide (D2O) as the primary deuterium source is reported. This method enables the direct deuteration of primary and secondary alcohols under basic or neutral conditions without being affected by coordinative functional groups such as imidazole and tetrazole. Successful substrates for deuterium labeling included losartan potassium, rapidosept, guaifenesin, and diprophylline. The deuterated losartan potassium showed higher stability towards the metabolism by CYP2C9 than the protiated analogue. Kinetic and DFT studies indicated that the direct deuteration proceeds through dehydrogenation of alcohol to the carbonyl intermediate, conversion of [Ir-III-H] to [Ir-III-D] with D2O, and deuteration of the carbonyl intermediate to give the alpha-deuterated product.

H/D Isotope Effects in Keto-Enol Tautomerism of β-Dicarbonyl Compounds -Importance of Nuclear Quantum Effects of Hydrogen Nuclei-

Summary: Deuterium isotope effects in the keto-enol tautomerism of beta-dicarbonyl compounds were studied using the B3LYP+D functional level of MC_DFT. The study showed that deuterium substitution decreases the keto-enol energy difference and weakens the intramolecular hydrogen-bonded interaction in the enol form. Direct treatment of NQEs of hydrogen nuclei via the MC_DFT method is crucial for analyzing the H/D isotope effect.
