Biochemical characterization and biological properties of mycelium extracts from Lepista sordida GMA-05 and Trametes hirsuta GMA-01: new mushroom strains isolated in Brazil

Guilherme Mauro Aranha et al.

Summary: This study evaluated the antioxidant activity, phenolic compounds, and cytotoxicity of mycelium extracts from two new Basidiomycete mushroom strains, Lepista sordida GMA-05 and Trametes hirsuta GMA-01, isolated in Brazil. The results showed that both mushroom extracts contained high amounts of proteins, free amino acids, carbohydrates, and phenolic compounds. The aqueous extract from T. hirsuta exhibited the highest antioxidant activity. Cytotoxicity tests on different cell lines revealed that L. sordida had higher toxicity in macrophages, while it was less toxic in Vero cells compared to T. hirsuta.

Supply chain logistics - the role of the Golgi complex in extracellular matrix production and maintenance

John Hellicar et al.

Summary: The biomechanical and biochemical properties of connective tissues are determined by the composition and quality of their extracellular matrix, which are highly influenced by the function and organization of the Golgi complex. Dysfunction of the Golgi has been found to have a significant impact on connective tissue in terms of disease and aging.

Synthesis, in vitro Antileishmanial Efficacy and Hit/Lead Identification of Nitrofurantoin-Triazole Hybrids

Nonkululeko H. Zuma et al.

Summary: Leishmaniasis is a common parasitic infection, especially prevalent among populations in low-income developing countries. The lack of effective vaccines means that chemotherapy is the primary treatment method. However, current antileishmanial drugs are limited due to their toxicity and drug-resistant strains. Therefore, there is a need for new and effective antileishmanial agents. In this study, a series of nitrofurantoin-triazole hybrids were synthesized and evaluated for their activity and cytotoxicity. One derivative, featuring a propargyl moiety, showed the highest activity against the causative pathogens of visceral leishmaniasis, with no toxicity to human cells. This compound has potential as an alternative therapy and warrants further investigation.

Summary: This study evaluated the in vitro activity of 18 benzohydrazones against M. tuberculosis, as well as their cytotoxicity, inhibition of M. tuberculosis efflux pumps, and ADMET assays. Some compounds showed to be possible efflux pump inhibitors, with most having high gastrointestinal absorption and ability to overcome the blood-brain barrier without irritant or tumorigenic effects. Compounds (1), (3), (9), (12), and (15) stood out for their good activities in both in vitro and in silico assays.
