Systematic Assessment of Uncoupling in Flavoprotein Oxidases and Monooxygenases

Summary: Flavin as a cofactor is involved in various biochemical reactions, and its unique ability to react with molecular oxygen in its reduced form sets it apart from other metal free cofactors. This study investigated the formation of reactive oxygen species (ROS) by several flavoprotein oxidases and monooxygenases, and found that significant amounts of superoxide were produced by all enzymes, while hydrogen peroxide levels were relatively low.

Biocatalytic Enantioselective β-Hydroxylation of Unactivated C-H Bonds in Aliphatic Carboxylic Acids

Summary: Selective hydroxylation of non-activated aliphatic C-H bonds was achieved through directed evolution of P450(BS beta) hydroxylase. The reaction showed high selectivity towards various aliphatic carboxylic acids with different chain lengths, functional groups, and aromatic moieties. The X-ray structure of the evolved enzyme variant provided insights into the regio- and enantioselectivity of the reaction.

Pilot-Scale Production of Peroxygenase from Agrocybe aegerita

Summary: The pilot-scale production of the peroxygenase from Agrocybe aegerita (rAaeUPO) is demonstrated, resulting in a final concentration of 0.29 g L-1 after 6 days of fermentation. However, only 170 g of the enzyme has been isolated due to nonoptimized downstream processing. The enzyme preparation showed preparative usefulness at a semipreparative scale for the stereoselective hydroxylation of ethyl benzene, achieving more than 340,000 catalytic turnovers and producing over 5 g L-1 (R)-1-phenyl ethanol in 7 days.
