Traditionally, protein design focused on active site residues for desired catalytic properties, but recent studies show that structural elements beyond the active site, like subunit-subunit interactions and flexible loops, also impact enzyme catalysis. This is particularly true for Rieske oxygenases, where the architectural features inside and outside the active site play a significant role in catalysis. Therefore, future Rieske oxygenase engineering efforts should consider these features.

Substrate-Specific Coupling of O2 Activation to Hydroxylations of Aromatic Compounds by Rieske Non-heme Iron Dioxygenases

Summary: Rieske dioxygenases play a crucial role in the initial steps of aromatic compound hydroxylation, but the mechanism behind O2 activation and substrate hydroxylation remains unclear. In this study, we focused on nitrobenzene dioxygenase and discovered that unproductive O2 activation is an important step in the catalytic cycle. By analyzing a series of substituted nitroaromatic compounds, we found that O2 uncoupling occurs after the formation of FeIII-(hydro)peroxo species, from which substrates are hydroxylated. Our findings provide a mechanistic basis for understanding the efficiency of substrate hydroxylation and unproductive O2 activation in reactions catalyzed by Rieske dioxygenases.

They use protons from water as terminal electron acceptors to dissipate excess reducing equivalents and produce hydrogen. FeFe-hydrogenases also have additional physiological functions, such as H-2 uptake, H-2 sensing, and CO2 fixation. Recent advancements in identifying and characterising novel FeFe-hydrogenases have expanded our understanding of their multiple roles and mechanisms.

Our findings showed that the binding site for Rieske-type Fd is commonly located at the stem (side-wise site) of the mushroom-like alpha(3)beta(3) quaternary structure in many ROs.

The constant development of tools and techniques has kept this field active and highly relevant in current research. This paper reviews the latest advancements in the understanding of the structural characteristics of nitrite reducing enzymes involved in both dissimilatory and assimilatory pathways.

Contrasting Mechanisms of Aromatic and Aryl-Methyl Substituent Hydroxylation by the Rieske Monooxygenase Salicylate 5-Hydroxylase

Summary: This study found that both hydroxylation and methyl hydroxylation of salicylate are catalyzed by monooxygenase chemistry. The functional unit involved in these reactions consists of a nonheme Fe(II) site and a Riesketype iron-sulfur cluster. Fluorine ring substituents were found to decrease the rate of Rieske electron transfer, suggesting a reaction between an Fe(III)-superoxo intermediate and the substrate. It was also discovered that both aromatic and aryl-methyl hydroxylation reactions start with the reaction of Fe(III)-superoxo with a ring carbon, but the latter must be reversible to yield a normal product isotope effect.
