Landscape and flux quantify the stochastic transition dynamics for p53 cell fate decision

Leijun Ye et al.

Summary: The study investigates the mechanisms of p53-regulated cell fate decisions using landscape and flux theory, quantifying potential landscape and probabilistic flux for the p53 system to explain the biphasic dynamics. In the oscillatory phase, landscape attracts the system into the ring valley and flux drives cyclic movement leading to cell cycle arrest; in the fate decision-making phase, the ring valley shape provides an efficient way for cells to return to the normal proliferative state after DNA damage repair. The analysis reveals a trade-off between stability (guaranteed by landscape) and function (driven by flux) in cellular systems.

Summary: By constructing a comprehensive gene regulatory network model and quantifying the potential landscape, it was found that EMT displays multiple stable attractors and the most probable transition paths were identified through the path-integral approach.
