P2X7 receptor: the regulator of glioma tumor development and survival

Damian Matysniak et al.

Summary: P2X7 receptor, known for its ionotropic nucleotide properties, has a wide range of effects on glioma biology, including promoting cell proliferation, enhancing cell adhesion, and influencing mitochondria depolarization and reactive oxygen species production. In vivo studies show that P2X7 activation in glioma tumors leads to tumor growth via pro-survival signaling pathways, but treatment with a selective antagonist inhibits tumor development and modulates the immune response. Upregulation of P2X7 receptor is also observed in human gliomas grades I to III, indicating its potential role in the disease progression.

Tumor Cell Infiltration into the Brain in Glioblastoma: From Mechanisms to Clinical Perspectives

Fidan Seker-Polat et al.

Summary: Glioblastoma is an extremely aggressive and invasive primary brain tumor with poor prognosis. Understanding the molecular mechanisms of glioblastoma invasion and developing new therapeutic approaches are of utmost importance. Efforts have been made to uncover the routes followed by glioblastoma cells, their interaction with the brain environment and extracellular matrix, and the role of cell adhesion and extracellular matrix remodeling. However, there is currently no approved therapy targeting the invasive phenotype.

P2X7 is a cytotoxic receptor horizontal ellipsis .maybe not: implications for cancer

Francesco Di Virgilio

Summary: The tumor microenvironment is rich in extracellular ATP, which affects cancer and immune cell responses through P2 receptors, particularly P2X7. The purinergic hypothesis proposed by Geoff Burnstock sparked innovative research in the field and provided a framework for interpreting numerous experimental findings. This appraisal highlights the influence of Geoff's inspiration on cancer studies and emphasizes the crucial role of the P2X7 receptor in investigations.

A small-molecule P2RX7 activator promotes anti-tumor immune responses and sensitizes lung tumor to immunotherapy

Laetitia Douguet et al.

Summary: A small subpopulation of non-small cell lung cancer patients respond to immunotherapies, and the development of HEI3090 as a chemical modulator of the purinergic P2RX7 receptor may enhance anti-tumor immune responses and sensitize lung cancer to PD-1 therapy in preclinical models.

Genetic Architectures and Cell-of-Origin in Glioblastoma

Hyun Jung Kim et al.

Summary: This study explores the pathogenesis and cell-of-origin of glioblastoma (GBM). It is suggested that GBM may originate from somatic mutations in neural stem cells and glial precursor cells. Further research will aid in understanding the evolution of GBMs and developing effective targets for treatment.

Characterization of Potency of the P2Y13 Receptor Agonists: A Meta-Analysis

Chrisanne Dsouza et al.

Summary: P2Y13 is an ADP-stimulated G-protein coupled receptor involved in various physiological processes. Understanding the activation dynamics of P2Y13 is crucial for translational studies. Differences in EC50 for ADP-like agonists were observed between brain and blood P2Y13, as well as between human and rodent P2Y13, suggesting potential tissue- and species-specific responses to ADP.

Structural basis for the functional properties of the P2X7 receptor for extracellular ATP

Lin-Hua Jiang et al.

Summary: The P2X7 receptor, originally known as the P2Z receptor, is a crucial member of the ATP-gated ion channel family, playing a significant role in ATP-induced purinergic signaling. Through functional characterizations and structural studies, we can gain valuable insights into the structure-function relationships of the P2X7 receptor.

Adenosine Inhibits Cell Proliferation Differently in Human Astrocytes and in Glioblastoma Cell Lines

Helena Marcelino et al.

Summary: The study found that the inhibitory effect of adenosine on glioblastoma multiforme (GBM) cells is significantly weakened, possibly due to increased adenosine elimination by adenosine kinase. Additionally, adenosine also enhances the cytotoxic effect of homocysteine on human astrocytes.
