Development of a highly specific and sensitive VHH-based sandwich immunoassay for the detection of the SARS-CoV-2 nucleoprotein

Marion Gransagne et al.

Summary: This study developed a sandwich immunoassay to specifically detect the nucleoprotein (N) of SARS-CoV-2. Seven antibodies were identified that recognized the full N protein and did not cross react with nucleoproteins from common human coronaviruses. The antibodies mainly targeted conformational epitopes in the C-terminal or N-terminal domains and were able to detect SARS-CoV-2 variants.

Structure and gating mechanism of the α7 nicotinic acetylcholine receptor

Colleen M. Noviello et al.

Summary: The alpha 7 nicotinic acetylcholine receptor plays critical roles in the central nervous system and the cholinergic inflammatory pathway as a ligand-gated ion channel. Recent cryo-EM structures of the human alpha 7 nicotinic receptor in different states have provided insights into its gating cycle and functional elements, including a C-terminal latch and an anionic ring that contribute to its high conductance and calcium permeability. Comparisons among the alpha 7 structures have revealed divergence in gating mechanisms within the Cys-loop receptor superfamily.

Therapeutic Targeting of α7 Nicotinic Acetylcholine Receptors

Roger L. Papke et al.

Summary: The alpha 7-type nicotinic acetylcholine receptor, with its unique characteristics and potential therapeutic importance, is the focus of research on subtype-selective agonists and conformational landscape. Challenges arise from rapid desensitization and ligand binding sites, but positive allosteric modulators and direct allosteric agonist or antagonists have diverse effects on receptor activation profiles.
