m6A modification: recent advances, anticancer targeted drug discovery and beyond

Li-Juan Deng et al.

Summary: Abnormal N6-methyladenosine (m6A) modification is closely related to cancer development, and finding targeted anticancer drugs is important. Traditional herbs and computer-synthesized compounds are reasonable sources for these drugs. Artificial intelligence has advantages in discovering m6A-targeting anticancer drugs.

METTL3 promotes intrahepatic cholangiocarcinoma progression by regulating IFIT2 expression in an m6A-YTHDF2-dependent manner

Qiong-Cong Xu et al.

Summary: This study reveals that METTL3 is upregulated in intrahepatic cholangiocarcinoma (ICC) and predicts poor prognosis. METTL3 expression is an independent predictor of overall survival in ICC patients. METTL3 can be transcriptionally activated by H3K4me3 and mediate m(6)A modification of IFIT2 mRNA, promoting ICC progression.

STAT3 promotes peritoneal metastasis of gastric cancer by enhancing mesothelial-mesenchymal transition

Hongkui Yang et al.

Summary: Activated STAT3 expression was found to be elevated in tumor tissues of gastric cancer patients, particularly those with peritoneal metastasis. Deactivating STAT3 suppressed proliferation, metastatic potential, and peritoneal metastasis of GC cells. These findings suggest that STAT3 activation plays a crucial role in promoting peritoneal metastasis of gastric cancer.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Small-molecule inhibition of METTL3 as a strategy against myeloid leukaemia

Eliza Yankova et al.

Summary: The study identified a highly potent and selective catalytic inhibitor of METTL3, STM2457, which shows potential therapeutic value in treating AML. Treatment with this inhibitor leads to reduced AML growth, increased differentiation and apoptosis, and decreased m(6)A levels on leukemogenic mRNAs. In vivo pharmacological inhibition of METTL3 results in impaired engraftment and prolonged survival in mouse models of AML.

NSD2 promotes tumor angiogenesis through methylating and activating STAT3 protein

Da Song et al.

Summary: The study showed that NSD2 promotes tumor angiogenesis through STAT3 mediation, with NSD2 enhancing STAT3 pathway activation by catalyzing STAT3 methylation; this reveals a potential NSD2-dependent methylation-phosphorylation regulation pattern of STAT3 in tumor angiogenesis.

FMRP regulates STAT3 mRNA localization to cellular protrusions and local translation to promote hepatocellular carcinoma metastasis

Zhifa Shen et al.

Summary: The study proposes a mechanism for the metastasis of hepatocellular carcinoma (HCC) cells by regulating the localization and translation of the STAT3 oncogene through fragile X mental retardation protein (FMRP). Knockdown of FMRP suppresses HCC metastasis effectively.

METTL3 promotes tumour development by decreasing APC expression mediated by APC mRNA N6-methyladenosine-dependent YTHDF binding

Wei Wang et al.

Summary: This study demonstrates that METTL3 downregulates APC by upregulating its m(6)A modification and recruiting YTHDF2 for APC mRNA degradation, leading to enhanced glycolysis and tumor growth in esophageal cancer.

Quantification of mRNA ribosomal engagement in human neurons using parallel translating ribosome affinity purification (TRAP) and RNA sequencing

Deivid Carvalho Rodrigues et al.

Summary: Translation regulation is a critical step in gene regulation, especially in neurodevelopment. The protocols described in the study involve calculating ribosomal engagement levels in in vitro-derived neuronal cells through enrichment of neurons, immunopurification of ribosome-bound RNAs, and calculation of ribosome-engaged mRNA fraction. The fraction of ribosome-engaged RNA serves as a measurement of translation activity, reflecting changes in translational regulation due to genotype or growth conditions.
