In Silico Design of Dual-Binding Site Anti-Cholinesterase Phytochemical Heterodimers as Treatment Options for Alzheimer's Disease

Summary: This study ranked phytochemicals with reported anti-acetylcholinesterase (AChE) activity based on in silico simulations and selected compounds with similar or higher binding affinity to AChE as galantamine. Novel dual-binding site heterodimeric drugs were designed using these compounds and showed potent inhibition of AChE and butyrylcholinesterase (BuChE) in computational experiments. The designed heterodimers displayed enhanced affinity to the target enzymes compared to the ChEI drug donepezil. In addition, the heterodimers showed drug-likeliness properties and low predicted toxicity. This study suggests that phytochemicals could be a valuable source for the development of novel ChEIs with improved efficacy and reduced toxicity.

Pharmacological blockade of the IL-6 signaling pathway has shown promising results in improving outcomes, especially when administered early in the disease course. Further research is needed to better understand the role of IL-6 in different COVID-19 phenotypes and identify patient subgroups that may benefit the most from IL-6 pathway inhibition.

Summary: The aqueous extract and biosynthesized nanoparticles of Manilkara zapota exhibit significant anti-inflammatory and analgesic activity against arthritis in rats. Nanoparticles show more pronounced antiarthritic effects compared to the aqueous extract, with free radical scavenging and reduction of TNF-alpha playing important roles in their activity. Further research is needed to identify the active phytochemical constituents responsible for the antiarthritic effects.
