Genome-wide CRISPR screen identifies CDK6 as a therapeutic target in adult T-cell leukemia/lymphoma

Takashi Ishio et al.

Summary: The study identifies CDK6 as a novel therapeutic target for adult T-cell leukemia/lymphoma (ATLL) and suggests the potential of combining CDK6 inhibitor palbociclib with mTORC1 inhibitors for treating this difficult malignancy.

CAMK1D Inhibits Glioma Through the PI3K/AKT/mTOR Signaling Pathway

Qianxu Jin et al.

Summary: CAMK1D is expressed at lower levels in glioma tissues and is positively correlated with the WHO classification of gliomas. Knockdown of CAMK1D promotes the proliferation, migration, and invasion of glioma cells, while overexpression has the opposite effect. Mechanistically, CAMK1D regulates glioma cell function through the PI3K/AKT/mTOR pathway. These findings suggest that CAMK1D may serve as a prognostic predictor and a potential therapeutic target for glioma.

Resistance to chemical carcinogenesis induction via a dampened inflammatory response in naked mole-rats

Kaori Oka et al.

Summary: Naked mole-rats exhibit resistance to cancer through a dampened inflammatory response. The study found that immune cell infiltration in the skin of naked mole-rats was significantly lower compared to mice after carcinogenic insults. The mole-rats carry mutations in two necroptosis-related genes, which may contribute to their enhanced cancer resistance.

Mutations in the miR-142 gene are not common in myeloproliferative neoplasms

Paulina Galka-Marciniak et al.

Summary: Recent data shows that MIR142 is one of the most frequently mutated miRNA genes and noncoding elements in all cancers, especially in blood cancers. Mutations in MIR142 have profound consequences for lympho- and myelopoiesis, and one of its targeted genes, CD274, is elevated in many cancer types, including MPNs. However, MIR142 mutations are rare in MPNs.

Suppression of PI3K/Akt/mTOR/c-Myc/mtp53 Positive Feedback Loop Induces Cell Cycle Arrest by Dual PI3K/mTOR Inhibitor PQR309 in Endometrial Cancer Cell Lines

I-Lun Hsin et al.

Summary: The study revealed a novel mechanism of the PI3K/mTOR dual inhibitor, PQR309, in lowering cell viability in endometrial cancer cells by disrupting the PI3K/Akt/mTOR/c-Myc/mtp53 positive feedback loop.
