Summary: Gastric cancer is the third leading cause of cancer-related deaths worldwide, with over 95% being adenocarcinomas. It is often diagnosed at an advanced stage, leading to a poor prognosis. However, systemic therapy can alleviate symptoms, improve survival, and enhance quality of life. The implementation of biomarker testing has significantly impacted clinical practice and patient care. Targeted therapies have shown encouraging results in clinical trials. Palliative management is recommended for patients with unresectable or metastatic cancer.

These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

The activation of the phosphatidylinositol-3-kinase (PI3K) axis in about one-third of human cancers, including gastric cancer, plays a significant role in the development of malignancies. This indicates the potential importance of targeting this axis in therapeutic strategies for such cancers.

Emetine shows potential as a promising drug candidate for chemotherapy of osteosarcoma.
