Effects of Ferroptosis on the Metabolome in Cardiac Cells: The Role of Glutaminolysis

Summary: In this study, the metabolome of H9c2 cardioblasts during RSL3-induced ferroptosis was elucidated using gas chromatography-mass spectrometry. The results showed that RSL3 decreased the levels of amino acids involved in glutathione synthesis and increased the levels of saturated fatty acids. RSL3 also significantly altered the levels of mitochondrial tricarboxylic acid cycle intermediates. Furthermore, ferrostatin-1, XJB-5-131, and TSM-1005-44 protected against RSL3-induced cell death and preserved the metabolomic profile of the cells.

Summary: Type 2 diabetic hearts are more susceptible to myocardial ischemia reperfusion (MIR) injury due to reduced mitophagy status. MitoQ, a mitochondria-targeted antioxidant, has been shown to protect the heart from ischemia reperfusion injury by upregulating mitophagy. In addition, MitoQ can alleviate cell damage caused by high glucose/high fat and hypoxia/reoxygenation, and improve cardiac function through the PINK1/Parkin pathway.

In contrast, mitochondrial-targeted antioxidants (MTAs) have emerged as a better alternative with higher advantages, but sufficient clinical trials have not been conducted to investigate their efficacy in PD treatment.

Summary: The purpose of this study was to evaluate the protective role of soursop leaves and avocado seed extract, single or in combination, against isoproterenol-induced myocardial infarction in male rats. The results showed that pretreatment with a combination extract of soursop leaf ethanol extract and avocado seeds significantly inhibited the increase of biomarker levels and maintained the integrity of the myocardial cell membrane.

Summary: This study observed the dynamic changes in myocardial mitochondrial damage and autophagic balance in rats treated with 5-FU, and found that 5-FU caused elevated myocardial enzyme levels, changes in cardiac structure and function, especially more severe in aging rats. Excessive mitochondrial autophagy induced by 5-FU worsened with longer administration time.
