Summary: Temozolomide (TMZ) is a DNA-methylating agent commonly used in cancer chemotherapy, particularly for glioblastoma multiforme (GBM). The study found that O(6)MeG, DNA double-strand breaks (DSB), and apoptosis all increase linearly with dose, while cellular senescence also shows a linear relationship without a threshold at therapeutically relevant dose levels. The genotoxic and cytotoxic effects observed suggest that the TMZ target concentration can trigger significant cytotoxic and cytostatic effects in vivo.
