Targeting the AnxA1/Fpr2/ALX pathway regulates neutrophil function, promoting thromboinflammation resolution in sickle cell disease

Junaid Ansari et al.

Summary: This study elucidated the functional significance of targeting the neutrophil-dependent AnxA1/FPR2/ALX pathway in SCD, showing that regulating the FPR2/ALX pathway improved cerebral thrombotic responses in Sickle transgenic mice and provided evidence of neutrophils' key role in thromboinflammation. Targeting the AnxA1/FPR2/ALX pathway may offer new therapeutic possibilities against thromboinflammatory conditions like SCD.

Differential pre-malignant programs and microenvironment chart distinct paths to malignancy in human colorectal polyps

Bob Chen et al.

Summary: Colorectal cancers originate from different precursor polyps, with factors related to stem cell expansion and cell differentiation playing key roles in tumor development; changes in the immune environment can also lead to mutations in tumor cells.

Human gastrointestinal epithelia of the esophagus, stomach, and duodenum resolved at single-cell resolution

Georg A. Busslinger et al.

Summary: The upper gastrointestinal tract, including the esophagus, stomach, and duodenum, controls different functions such as food transport, digestion, and hormone production. Through single-cell analysis, distinct cell types have been identified in these organs, with specific populations like quiescent stem/progenitor cells and rare BCHE cells in the esophagus and duodenum respectively. Gene expression varies significantly between identical cell types in different organs, and there are unique cell types like serotonin-producing enterochromaffin cells and histamine-producing enterochromaffin-like cells that express additional hormones such as luteinizing hormone.

Integrated single-cell transcriptome analysis reveals heterogeneity of esophageal squamous cell carcinoma microenvironment

Huy Q. Dinh et al.

Summary: This study utilized single-cell RNA sequencing to characterize the heterogeneous microenvironment of ESCC, identifying a tumor-specific subset of CST1(+) myofibroblasts with prognostic values and immunosuppressive features. The analysis of stromal cell types in the ESCC microenvironment provides a rich resource for further understanding of ESCC biology.

Role of Annexin A1 in Squamous Cell Lung Cancer Progression

Omar Elakad et al.

Summary: In this study, we investigated ANXA1 expression in patient-derived tissue samples of lung cancer and found that strong ANXA1 expression was significantly correlated with longer overall survival in SQCLC patients. Mechanistically, ANXA1 overexpression promoted proliferation but suppressed migration in SQCLC cell lines, while knockout of ANXA1 had the opposite effects. These findings suggest that ANXA1 expression may contribute to longer survival by inhibiting tumor cell migration and metastasis.

Cancer-associated fibroblasts activated by miR-196a promote the migration and invasion of lung cancer cells

Sieun Lee et al.

Summary: This study reveals that miR-196a promotes the migration and invasion of lung cancer cells by regulating ANXA1 and CCL2 in the tumor microenvironment. High expression of miR-196a is associated with poor prognosis in lung adenocarcinoma patients.

Integrated analysis of multimodal single-cell data

Yuhan Hao et al.

Summary: The study introduces a weighted-nearest neighbor analysis framework to learn the relative utility of each data type in each cell, enabling integrative analysis of multiple modalities. Applied to a CITE-seq dataset, the method constructs a multimodal reference atlas of the circulating immune system and successfully identifies and validates previously unreported lymphoid subpopulations.

LAMC1 upregulation via TGFβ induces inflammatory cancer-associated fibroblasts in esophageal squamous cell carcinoma via NF-κB-CXCL1-STAT3

Lingling Fang et al.

Summary: This study identified a dual mechanism by which TGF beta upregulates LAMC1 in tumor cells, promoting ESCC proliferation and migration, as well as indirectly inducing carcinogenesis by stimulating CXCL1 secretion to promote the formation of inflammatory CAF. This finding suggests that LAMC1 could be a potential therapeutic target and prognostic marker for ESCC.
