Myeloid PTEN deficiency aggravates renal inflammation and fibrosis in angiotensin II-induced hypertension

Changlong An et al.

Summary: The study investigated the role of PTEN in renal inflammation and fibrosis induced by hypertension, showing that PTEN plays a crucial role in the pathogenesis of renal inflammation and fibrosis by regulating the infiltration of myeloid fibroblasts, macrophages, and T lymphocytes into the kidney.

STAT6 Deficiency Attenuates Myeloid Fibroblast Activation and Macrophage Polarization in Experimental Folic Acid Nephropathy

Baihai Jiao et al.

Summary: STAT6 plays a critical role in myeloid fibroblast activation, M2 macrophage polarization, extracellular matrix protein production, and renal fibrosis development in folic acid nephropathy. Knockout of STAT6 results in reduced fibroblast numbers, decreased M2 macrophage polarization, and less severe interstitial fibrosis in the kidney with folic acid nephropathy. Targeting STAT6 may offer a novel therapeutic strategy for fibrotic kidney disease.

Inhibition of macrophage histone demethylase JMJD3 protects against abdominal aortic aneurysms

Frank M. Davis et al.

Summary: JMJD3 plays a critical role in the pathogenesis of abdominal aortic aneurysms by regulating the inflammatory immune response in macrophages. Targeted inhibition of JMJD3 can reduce AAA expansion and attenuate macrophage-mediated inflammation, suggesting that cell-specific pharmacologic therapy targeting JMJD3 may be an effective intervention for AAA.

Pharmacological Inhibition of STAT6 Ameliorates Myeloid Fibroblast Activation and Alternative Macrophage Polarization in Renal Fibrosis

Baihai Jiao et al.

Summary: The study demonstrated that the selective STAT6 inhibitor, AS1517499, effectively inhibits STAT6 activation, reduces the accumulation of myeloid fibroblasts and myofibroblasts in the kidney in experimental models of unilateral ureteral obstruction or folic acid nephropathy, suppresses M2 macrophage polarization, attenuates extracellular matrix protein production, and preserves kidney function. Targeting STAT6 with AS1517499 represents a novel therapeutic approach for chronic kidney disease.
