Circulating cell-free tumor human papillomavirus DNA is a promising biomarker in cervical cancer

Summary: This study aimed to analyze the detectability of cell-free human papillomavirus DNA (ctHPV DNA) in patients with cervical cancer or pre-malignant lesions and its correlation with patient and tumor characteristics and outcome. The results showed a high proportion of ctHPV DNA detection in patients with locally advanced cervical cancer, and its levels were correlated with disease staging. Furthermore, the presence of ctHPV DNA was associated with worse progression-free survival in patients.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

cfDNA detection for HPV plus squamous cell carcinomas

Summary: High-risk HPV is a factor in various squamous cell carcinomas and its cfDNA levels can be indicative of poor prognosis and treatment relapse. Longitudinal sampling has shown that persistent or increasing HPV cfDNA levels can predict treatment failures. Using a liquid biopsy platform for HPV cfDNA may have clinical utility in HPV+ cancer patients.

This suggests the importance of predicting the functional consequences of FGFR2 mutations for guiding treatment decisions in iCCA patients.
