Global Genomic and Proteomic Analysis Identified Critical Pathways Modulated by Proto-Oncogene PELP1 in TNBC

Summary: In this study, the molecular mechanisms by which PELP1 contributes to the progression of TNBC were elucidated using proteomic and RNA-seq approaches. The results showed that PELP1 regulates the expression of ribosomal proteins and ribosomal regulatory complexes, as well as the functions of transcription factor c-Myc in TNBC. These findings have important implications for understanding the pathological mechanisms of TNBC.

A First-in-Class Inhibitor of ER Coregulator PELP1 Targets ER plus Breast Cancer

Summary: A novel peptide inhibitor called SMIP34 has been found to efficiently inhibit the oncogenic signaling of PELP1, a coregulator implicated in therapy-resistant ER+ breast cancer. SMIP34 blocks the interaction between PELP1 and its target, reduces cell growth, induces PELP1 degradation, and alters gene expression related to estrogen response, cell cycle, and apoptosis pathways.

Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.
