Brain GLP-1 and the regulation of food intake: GLP-1 action in the brain and its implications for GLP-1 receptor agonists in obesity treatment

Stefan Trapp et al.

Summary: This review examines the similarities and differences between physiological systems regulated by gut-derived and neuronally produced GLP-1, discussing whether peripheral and central sources of GLP-1 are separate, linked, or redundant systems, as well as the heterogeneity or homogeneity of the brain GLP-1 system, and the implications for using GLP-1 receptor agonists as anti-obesity drugs.

Actions of glucagon-like peptide-1 receptor ligands in the gut

Jens Juul Holst et al.

Summary: GLP-1, inactivated by DPP-4 before leaving the gut, likely acts through activation of intestinal sensory neurons expressing its receptors. Its effects on appetite, gastrointestinal secretion and motility, and pancreatic secretion are probably mediated by activation of vagal afferents. Additionally, GLP-1 receptors are widely expressed throughout the GI tract, indicating potential local effects on epithelial protection, endocrine signaling, and interaction with the immune system.

Oxytocin acts centrally in the brain to improve leaky gut through the vagus nerve and a cannabinoid signaling in rats

Toshikatsu Okumura et al.

Summary: Brain oxytocin plays an important role in regulating intestinal barrier function. It can reduce colonic hyperpermeability and this effect is mediated by the vagal cholinergic pathway or cannabinoid 1 receptor signaling. This finding suggests that activation of central oxytocin signaling may be beneficial for the treatment of leaky gut-related diseases such as irritable bowel syndrome and autism.

Distribution and ultrastructural localization of the glucagon-like peptide-1 receptor (GLP-1R) in the rat brain

Erzsebet Farkas et al.

Summary: GLP-1R protein is mainly distributed in circumventricular organs and neighboring regions such as the arcuate nucleus and nucleus tractus solitarii in the rat brain. Additionally, GLP-1R protein is also present in various areas of the telencephalon, diencephalon, brainstem, and cerebellum. Ultrastructural examination reveals that GLP-1R immunoreactivity is associated with the membrane of perikarya and dendrites, and can also be found inside and on the surface of axon varicosities and terminals.

Phlorizin attenuates visceral hypersensitivity and colonic hyperpermeability in a rat model of irritable bowel syndrome

Tsukasa Nozu et al.

Summary: Phlorizin was found to improve visceral hypersensitivity and colonic hyperpermeability in IBS models by inhibiting SGLT2, and these effects were mediated via the AMPK, NO, and opioid pathways.

Activation of central adenosine A2B receptors mediate brain ghrelin-induced improvement of intestinal barrier function through the vagus nerve in rats

Masatomo Ishioh et al.

Summary: Through activating adenosine A2B receptors in the central nervous system, intestinal barrier function can be improved; these receptors may mediate the ghrelin-induced improvement of leaky gut in a vagal dependent fashion, which helps us understand the pathophysiology not only in gastrointestinal diseases but also in non-gastrointestinal diseases associated with altered intestinal permeability.

Central and peripheral GLP-1 systems independently suppress eating

Daniel I. Brierley et al.

Summary: GLP-1 suppresses eating through independent gut-brain circuits, with PPG(NTS) neurons not being necessary for the eating suppression effects of GLP-1 receptor agonists. Concurrent activation of PPG(NTS) neurons can more effectively suppress eating than using semaglutide alone.
