High-throughput functional analysis of autism genes in zebrafish identifies convergence in dopaminergic and neuroimmune pathways

Hellen Weinschutz Mendes et al.

Summary: This study performs parallel in vivo functional analysis of 10 ASD genes at the behavioral, structural, and circuit levels in zebrafish mutants, revealing both unique and overlapping effects of gene loss of function. Whole-brain mapping identifies the forebrain and cerebellum as the most significant contributors to brain size differences, while regions involved in sensory-motor control, particularly dopaminergic regions, are associated with altered baseline brain activity. Finally, a global increase in microglia resulting from ASD gene loss of function in select mutants implicates neuroimmune dysfunction as a key pathway relevant to ASD biology.

Behavioral and Sensory Deficits Associated with Dysfunction of GABAergic System in a Novel shank2-Deficient Zebrafish Model

Yi Wang et al.

Summary: Hyper-reactivity to sensory inputs is a common symptom of autism spectrum disorder (ASD), but its underlying neural abnormalities are unclear. By studying a zebrafish model with SHANK2 mutations, the study found that the deficiency of SHANK2 contributes to sensory abnormalities and ASD-like behaviors. The model exhibited reduced social preference, enhanced behavioral stereotypy, hyper-sensitivity to auditory noise, and abnormal hyperactivity. The study also identified GABAergic dysfunction as a major contributor to sensory hyper-reactivity in ASD, emphasizing the importance of early interventions to prevent disease progression.

Transplantation of high fat fed mouse microbiota into zebrafish larvae identifies MyD88-dependent acceleration of hyperlipidaemia by Gram-positive cell wall components

Pradeep Manuneedhi Cholan et al.

Summary: The study demonstrates that high fat-diet-associated gut microbiota can increase susceptibility to hyperlipidaemia in zebrafish larvae, driven by two individual bacterial species. Specific bacteria such as Stenotrophomonas maltophilia and Enterococcus faecalis contribute to the hyperlipidaemic effect, independent of direct interaction with the host. Additionally, toll-like receptor agonists peptidoglycan and lipoteichoic acid can replicate the hyperlipidaemic effect through a MyD88-dependent mechanism.

Immune Dysregulation in Autism Spectrum Disorder: What Do We Know about It?

Maria de los Angeles Robinson-Agramonte et al.

Summary: Autism Spectrum Disorder (ASD) is a complex neurodevelopmental disorder characterized by deficits in social communication, restricted interests, and repetitive behavior. The immune hypothesis plays a major role in ASD pathogenesis and explains the variability in clinical phenotypes and comorbidities. Evidence suggests a link between immune dysfunction and behavioral traits in ASD. This review summarizes current understanding of immune dysfunction in ASD, particularly the impact of maternal immune factors, comorbidities, and other relevant factors.

Short chain fatty acids: Microbial metabolites for gut-brain axis signalling

Kenneth J. O'Riordan et al.

Summary: The role of the intestinal microbiota in regulating gut-brain axis signaling has gained attention in recent years. Understanding the relationship between the gut microbiota, its metabolites, and the brain is crucial for developing new therapeutic approaches, including identifying psychobiotics. Current research focuses on the effects of short-chain fatty acids (SCFAs), such as butyrate, acetate, and propionate, on brain function, behavior, and physiology, and how these SCFAs enter the systemic circulation and can potentially impact the brain.

Loss of autism-candidate CHD8 perturbs neural crest development and intestinal homeostatic balance

Gaelle Hayot et al.

Summary: In this study, using a chd8 mutant zebrafish model, researchers found that loss of chd8 led to reduced numbers of vagal neural crest cells, as well as altered early migration capability. Furthermore, they observed decreased numbers of serotonin-producing enterochromaffin cells and NCC-derived serotonergic neurons, suggesting intestinal hyposerotonemia in the absence of chd8. Transcriptomic analyses revealed altered expression of key receptors and enzymes in serotonin and acetylcholine signaling pathways. Tissue examination of chd8 mutants showed a thinner intestinal epithelium, accumulation of neutrophils, decreased numbers of goblet cells and eosinophils. Additionally, single-cell sequencing demonstrated a disruption of immune balance with changes in inflammatory interleukins and immune cell clusters.

Summary: This study demonstrates that epithelial sensory enteroendocrine cells (EEC) in the intestinal epithelium can sense and respond to microbial stimuli through the Trpa1 receptor, leading to activation of vagal neuronal pathways and increased intestinal motility. Furthermore, indole derivatives of tryptophan catabolism also play a role in this process by activating Trpa1 signaling and stimulating intestinal serotonin secretion.
