Increasing precision in the management of pediatric neurosurgical cerebrovascular diseases with molecular genetics

Kristopher T. Kahle et al.

Summary: Recent sequencing studies have revealed new disease genes and signaling pathways involved in congenital and pediatric cerebrovascular anomalies, providing insights into the genetic regulation of human cerebrovascular development. By classifying these disorders based on molecular disruptions in signaling pathways, it is possible to replace the current anatomically based disease classification systems and improve precision in genetic counseling, prognosis, and treatment stratification. Moreover, molecular genetic data are already influencing clinical trials and the development of targeted therapies for these conditions.

The Genetic Basis of Moyamoya Disease

R. Mertens et al.

Summary: Moyamoya disease is a rare cerebrovascular disease characterized by progressive bilateral occlusion of the intracranial internal cerebral arteries. Clinical presentation varies depending on age and population, with hemorrhage and ischemic infarcts leading to severe neurological dysfunction or death. Surgical revascularization is currently the only established treatment for MMD.

Ca2+-activated Cl- channel TMEM16A inhibition by cholesterol promotes angiogenesis in endothelial cells

Ke Ma et al.

Summary: This study reveals a novel mechanism of cholesterol-mediated TMEM16A inhibition, by which cholesterol reduces TMEM16A expression via DNMT1-mediated methylation and directly inhibits channel activities. TMEM16A channel inhibition promotes endothelial cell angiogenesis.
