Discovery of Novel Activators of Large-Conductance Calcium-Activated Potassium Channels for the Treatment of Cerebellar Ataxia

Sharan R. Srinivasan et al.

Summary: Impaired firing of cerebellar Purkinje neurons due to reduced expression of BK channels is a common feature in inherited neuro-degenerative spinocerebellar ataxia (SCA). This study successfully identified a novel BK channel activator that improved the irregular firing patterns of SCA1 Purkinje neurons.

Afterhyperpolarization Promotes the Firing of Mitral Cells through a Voltage-Dependent Modification of Action Potential Threshold

Nicolas Fourcaud-Trocme et al.

Summary: In the olfactory bulb, mitral cells (MCs) exhibit spontaneous bursting activity, characterized by bursts of action potentials (APs) intermixed with silent periods. The burst initiation threshold and duration are dependent on the membrane potential, which in turn is regulated by the inactivation state of voltage-dependent Na+ channels. The fast afterhyperpolarization (AHP) promotes burst firing, while the slow AHP contributes to burst termination.

Modulation of the CB1 cannabinoid receptor has potential therapeutic utility in the 3-acetylpyridine cerebellar ataxia rat model

Hoda Ranjbar et al.

Summary: This study suggests that cannabinoid agonists, specifically the CB1R agonist WIN, may not be effective in treating cerebellar ataxia and may even worsen symptoms. However, co-administration of the CB1R antagonist AM251 with WIN can prevent some ataxic symptoms. This indicates that cannabinoid modulation of ataxia symptoms may involve other cannabinoid receptors in addition to CB1Rs.

The neglected role of endocannabinoid actions at TRPC channels in ataxia

Hoda Ranjbar et al.

Summary: TRP channels play a crucial role in the pathogenesis of ataxia, with their activity modulated by endogenous endocannabinoids. Targeting TRP channels, particularly TRPC channels, may offer a potential therapeutic approach for the management of ataxia.

Potassium channels as molecular targets of endocannabinoids

Yu-Fung Lin

Summary: Endocannabinoids are endogenous mediators derived from membrane lipids, influencing various physiological functions through activation of different receptors and channels, including potassium channels. They not only regulate action potentials and cell excitability, but also hold potential therapeutic value.
