Revising the JBI quantitative critical appraisal tools to improve their applicability: an overview of methods and the development process

Timothy Hugh Barker et al.

Summary: JBI provides critical appraisal instruments for systematic reviewers and researchers to investigate methodological limitations in primary research studies. These study-specific instruments are presented as checklist-style questions. However, many of the tools offered by JBI have become outdated as the field of research synthesis expands, necessitating updates.

Impaired TRPM3-dependent calcium influx and restoration using Naltrexone in natural killer cells of myalgic encephalomyelitis/chronic fatigue syndrome patients

Natalie Eaton-Fitch et al.

Summary: This study found that the calcium influx in NK cells of ME/CFS patients was impaired. However, the overnight treatment of isolated NK cells with NTX restored the TRPM3-dependent calcium influx. These findings suggest that ME/CFS may be a TRP ion channel disorder and that NTX could be a potential therapeutic intervention.

Endothelial dysfunction and altered endothelial biomarkers in patients with post-COVID-19 syndrome and chronic fatigue syndrome (ME/CFS)

Comparative Analysis of Extracellular Vesicles in Patients with Severe and Mild Myalgic Encephalomyelitis/Chronic Fatigue Syndrome

Summary: Myalgic encephalomyelitis, or chronic fatigue syndrome (ME/CFS), is a serious disease with an unknown cause and objective markers. This study found potential dysregulation of B cell and platelet activation or homeostasis in ME/CFS patients, which could be important for diagnosis and treatment.

miR-21: a non-specific biomarker of all maladies

Ana E. Jenike et al.

Summary: miRNA-21 is considered as a specific predictive or prognostic biomarker for at least 29 diseases, but lacks specificity for any one disease, thus making it not an ideal candidate for a biomarker despite continued evaluation. This issue of non-specificity in biomarker designation is shared with other common, ubiquitous miRNAs and warrants concern for their use as biomarkers as well.

Submaximal Exercise Provokes Increased Activation of the Anterior Default Mode Network During the Resting State as a Biomarker of Postexertional Malaise in Myalgic Encephalomyelitis/Chronic Fatigue Syndrome

Rakib U. Rayhan et al.

Summary: This study found that individuals with ME/CFS showed increased spontaneous activity in the medial prefrontal cortex, the anterior node of the Default Mode Network, after exercise, while controls exhibited decreased activation in this region. Overall, controls had higher BOLD signals indicating reduced global cerebral blood flow in ME/CFS.

Markers of Cardiac Autonomic Function During Consecutive Day Peak Exercise Tests in People With Myalgic Encephalomyelitis/Chronic Fatigue Syndrome

Maximillian J. Nelson et al.

Summary: Patients with ME/CFS exhibit altered ventilatory characteristics and higher HR parameters on the second day of consecutive CPET tests, suggesting potential physiological markers. Lower post-exercise HR recovery was identified in ME/CFS patients. However, the limited sensitivity and specificity of HRR values restrict their potential usefulness as a biomarker for the condition.

Tryptophan Metabolites, Cytokines, and Fatty Acid Binding Protein 2 in Myalgic Encephalomyelitis/Chronic Fatigue Syndrome

Manuela Simonato et al.

Summary: Patients with ME/CFS exhibit heterogeneity in clinical and biochemical traits, with higher levels of IL-17A, FABP-2, and 3-hydroxykynurenine and lower levels of kynurenine and serotonin compared to healthy controls. The changes in tryptophan metabolites and increased IL-17A levels in ME/CFS may be associated with anomalies in energy metabolism. Further research on serum biomarkers related to inflammation, intestine function, and tryptophan metabolism is warranted for personalized medicine strategies for ME/CFS.

Activin B levels tended to decrease with increasing visual analogue scale score, but the results do not support its clinical utility as a biomarker for ME/CFS at this time.
