Leveraging Northern European population history: novel low-frequency variants for polycystic ovary syndrome

Summary: By leveraging the unique population history of Northern Europe, we have identified three novel genome-wide significant associations with polycystic ovary syndrome (PCOS), including two putative independent causal variants in the CHEK2 gene and one variant near the MYO10 gene. This study encourages the use of isolated populations to uncover rare variants contributing to the genetic landscape of complex diseases like PCOS.

Identifying molecular mediators of the relationship between body mass index and endometrial cancer risk: a Mendelian randomization analysis

Summary: This study used Mendelian randomization to investigate the causal role of molecular risk factors in endometrial cancer risk, and to evaluate the potential mediating role of these factors in the relationship between body mass index (BMI) and endometrial cancer. The results showed that BMI, testosterone, bioavailable testosterone, fasting insulin, and sex hormone-binding globulin had a causal effect on endometrial cancer risk. Additionally, total serum cholesterol may also have an effect on endometrial cancer risk. Insulin, testosterone, and sex hormone-binding globulin were found to mediate the relationship between BMI and endometrial cancer risk.

Measuring the global disease burden of polycystic ovary syndrome in 194 countries: Global Burden of Disease Study 2017

Summary: The study revealed slight increases in age-standardized incidence of PCOS and associated disability-adjusted life-years (DALYs) among women of reproductive age globally from 2007 to 2017, especially in middle-SDI and high-middle SDI regions. Understanding the differences in the burden of PCOS across various locations could be valuable for resource allocation and formulation of effective preventive strategies.
