Rotenone-Induced Neurodegeneration Is Enabled by a p38-Parkin-ROS Signaling Feedback Loop

Jialong Chen et al.

Summary: The study revealed a novel feedback mechanism of p38-Parkin-ROS regulating rotenone-induced Parkinson's disease. Both p38 activation and Parkin inactivation contribute to the disruption of Parkin-mediated mitophagy, leading to increased reactive oxygen species (ROS) production. Inhibiting p38 and enhancing Parkin activity can reduce ROS levels and restore cell death and mitochondrial function in rotenone-induced PD models.

Irx3 and Irx5-Novel Regulatory Factors of Postnatal Hypothalamic Neurogenesis

Zhengchao Dou et al.

Summary: The hypothalamus, a highly conserved brain region, plays a crucial role in regulating physiological processes like energy balance. Neurons in the arcuate nucleus sense peripheral signals and regulate food intake and energy expenditure. Postnatal neurogenesis in the mouse hypothalamus, particularly in the first two postnatal weeks, contributes to remodeling and energy balance regulation. Radial glia-like cells, including RGL-NSCs, play a key role in this neurogenesis process, with genetic factors such as Irx3 and Irx5 influencing their properties. Understanding these mechanisms provides insights into hypothalamic remodeling and energy balance control.

MiR-421 Aggravates Neurotoxicity and Promotes Cell Death in Parkinson's Disease Models by Directly Targeting MEF2D

Yaru Dong et al.

Summary: Parkinson's disease is characterized by selective loss of dopaminergic neurons, and studies have shown that miR-421 and MEF2D are dysregulated in a cellular model of PD, with miR-421 promoting cell death through negative modulation of MEF2D expression. Inhibition of miR-421 or restoration of MEF2D protects neurons from neurotoxicity in models of PD, highlighting a potential novel mechanism in the pathogenesis of the disease.
