DNA-Protein Crosslinks and Their Resolution

Summary: This review provides an overview of the sources and principles of cellular DPC formation, as well as the cellular mechanisms resolving these threats to genomic integrity. Repair of DPCs requires the action of canonical DNA repair pathways and specialized proteolytic enzymes. Loss of DPC repair capacity has dramatic consequences.

Topoisomerase I inhibitors: Challenges, progress and the road ahead

FDA-approved CPT derivatives have limitations, leading to interest in developing non-CPT Top1 poisons. Summary: Defects in DNA double-strand break repair, such as BRCA1 or BRCA2 mutations, make tumors selectively sensitive to PARP inhibitors. However, BRCA and PARP1 also share functions in DNA synthesis on the lagging strand, suggesting that BRCA deficiency may be due to an inherent problem in lagging strand synthesis.

TDP1-independent pathways in the process and repair of TOP1-induced DNA damage

Summary: The authors found that MUS81 plays a role in mediating excess DNA double-strand break (DSB) generation in TDP1 KO cells after camptothecin treatment. They demonstrate that TOP1 cleavage complexes can either be directly resolved by TDP1 or be converted into DSBs and repaired by the Homologous Recombination pathway.
